Abstract: The effects of ionomycin dealing time, ionomycin dealing density, different basic embryo culture mediums on parthenogenetic activation of porcine zona-free oocytes were studied. The results are as follows: (1) The activation rate with 10 μM ionomycin dealing 5 min(64.81%) was not significantly different from that of dealing 7 min(66.67%), 10 min(68.75%), but significantly higher than 3 min(36.17%). (2) The activation rate of ionomycin dealing 10 min with 10μM(68.18%) was not significantly different from that with 15μM (65.11%), but significantly higher than that with 5μM (40.54%). (3) The activation rate of using NCSU-23+4%BSA and TCM-199+4%BSA as embryo basic culture mediums was not significantly different(68.51%, 62.50%). The results of this experiment showed that 10 μM ionomycin dealing 5 min in combination with using NCSU-23+4%BSA can effectively activate porcine zona-free oocytes.
