  Effcets of Insulin-Like Growth Factor 1 on the Proliferation at Different Generation of Antler Organic Center Cells Mei Mei, Deng Xuming, Yue Zhanpeng, Zhao Lihong, Li Chunyi, Feng Haihua 2008, 24 (4):  1-3.  Abstract + ( 2107 )   PDF ( 374209KB )   ( 1503 ) 【Objective】To investigate the effects of insulin-like growth factor 1 (IGF1)on the proliferation of antler organic center cells from various passages in vitro at 60 days growing period of antler. 【Method】Antler organic center cells were isolated from sika deer antler of growing 60 day and cultured. The second, fifth and eighth population cells were incubated with 0, 1, 3, and 10 nM IGF1. Thymidine incorporation assay was applied after 24 hours of culture respectively to determine the disintegration per minute ( DPM) value.【Results】The proliferation of cells from growing 60 day’s antler in the experimental groupwas significantly higher than that of the control group (p<0.01). The average and maximal DPM value of the proliferation of cells in the experimental group was the highest in the second population cells (31918 and 39818 DPM/mg respectively), decreased in the fifth population cells materially (5455 and 6815 DPM/mg), and decreased in the eighth population cells again (4030 and 4838 DPM/mg). 【Conclusion】The results show that IGF1 can enhance the proliferation of antler organic center cells cultured in vitro. The sensitivity of IGF1 on the antler organic center cells from various passages at 60 days growing period of antler in vitro.

The results showed indigowoad root polysaccharides in the cell concentration of security, porcine parvovirus obviously blocking role and the role of inhibition, the smallest concentration of 2.09μg/ml block, minimum inhibitory concentration for 4.19μg/ml.

N gene’s cDNA fragment of about 1.6kb was obtained through RT-PCR from FOX TA strain of CDV, and was cloned into pIREShyg Vector Eukarytic Expression Vector named as pIRES-N was constructed. pIRES-N was transfected into CHO-K1 cells with coprecipitation, and positive cells were screened with hygromycine selection. The N gene protein was expressed in the CHO cells proved by IFA. The N gene was transcripted as proved with RT-PCR. CHO/CDV-N cell strain was constructed, which provided evidence for research of CDV serumal detection and gene vaccine.

  Studies on Different Methods of Parthenogenetic Activation on Porcine Zona-free Oocytes Hu Junhe , , Qu Lei, , Jing Xiaoqi , 2008, 24 (4):  13-15.  Abstract + ( 1960 )   PDF ( 390125KB )   ( 1077 ) The effects of ionomycin dealing time, ionomycin dealing density, different basic embryo culture mediums on parthenogenetic activation of porcine zona-free oocytes were studied. The results are as follows: (1) The activation rate with 10 μM ionomycin dealing 5 min(64.81%) was not significantly different from that of dealing 7 min(66.67%), 10 min(68.75%), but significantly higher than 3 min(36.17%). (2) The activation rate of ionomycin dealing 10 min with 10μM(68.18%) was not significantly different from that with 15μM (65.11%), but significantly higher than that with 5μM (40.54%). (3) The activation rate of using NCSU-23+4%BSA and TCM-199+4%BSA as embryo basic culture mediums was not significantly different(68.51%, 62.50%). The results of this experiment showed that 10 μM ionomycin dealing 5 min in combination with using NCSU-23+4%BSA can effectively activate porcine zona-free oocytes.

The results show that the method of freezing-centrifugation is more effective than that of other methods, the recovery rate is high, sericin powder prepared by ethanol precipitation possesses excellent physical property, its solubility and absorbability to water and oil are higher than that of other methods, it exhibits preferable functional property.

  Effects of FMDV 146S on IL-6 Release of Suckling Rat Myocardial Microvascular Endothelial Cells in Vitro Tian Lifang, Zhang Tao, Zhang Shuang , Chen Wu, Suo Zhanwei, Hu Ge, Duan Huiqin, Mu Xiang 2008, 24 (4):  19-22.  Abstract + ( 1831 )   PDF ( 496191KB )   ( 1012 ) To study the effects foot and mouth diseases virus 146S particle antigen (FMDV 146S) on IL-6 release of sucking rat myocardial microvascular endothelial cells (RMMVECs) in vitro, and to provide data for solving the problem of side effects in using foot-and-mouth-disease vaccines. The cultured RMMVECs in vitro were identified as endothelial cells by morphology and by positive immunocytochemistry for factor Ⅷ-related antigen, a marker for endothelial cells. Using enzyme linked immunosorbent assay (ELISA) to detect the level of IL-6 produced by RMMVECs stimulated with FMDV 146S in cell supernatant liquid. On normal condition, RMMVECs can secrete IL-6 at low level. The level of IL-6 produced by RMMVECs stimulated with FMDV146S in cell supernatant liquid was increased dramatically in dose and time dependents, and got the maximum at 24 h. MVECs are very significant in immune and inflammatory reaction induced by FMDV 146S.

The quality of RNA isolated from medicinal plants was usually poor and was not ideal as start material for molecular research. In order to solve this problem, one kind of RNA extraction method which was suitable for different tissues of Rehmannia glutinosa was explored and the RNA quality was tested by subsequent molecular experiments. The results showed that the extracted RNA isolated by this method was pure and integral and could satisfy molecular experiment needs. It also showed that the method can be used for RNA extraction of other medicinal plants.

  An Affection Method for DNA Extraction from Soil Microorganisms Chen Xuyu, Zhou Yakui, Yu Xianmei, Zheng Fucong, 2008, 24 (4):  33-36.  Abstract + ( 1967 )   PDF ( 859258KB )   ( 1285 ) In this paper we test with soil microorganisms from Hevea forest, and direct extract microbial nucleic acid from soil, The research shows that this method of introduction in this experiment not only generate the largest DNA fragments in size, but also it was directly used as PCR amplification and digestion of restriction enzymes and later operation without purification, about 2.1-4.6μg DNA yields could be extracted from one gram soil on average. This method is easy and quickly in operation, and provide the basis of the construction of metagenomic library and diversity analysis of soil microbial communities.

Optimized SSR system for peanut was established : 25 mM MgCl2 1.5 μl, 2.5 mM dNTP 1.6 μl, 5 U/μl Taq 0.17 μl, 100 ng/μl template DNA 0.4 μl, 2.5 mΜ primers 5 μl were added to the PCR mixture. Optimized annealing temperature was 54℃.

Plasmid pEGFP and pCAMBIA1300 were digested by Hind III and EcoRⅠ restriction enzymes and ligated to transform. We constructed Tremella fuciformis expressed vector pCB-BEGFP contained enhanced green fluorescent protein gene which was regulated by gpd promoter of Agaricus bisporus. Electroporation was performed to transfer plasmid DNA of pCB-BEGFP into yeast-like conidia from Tremella fuciformis. The chromosomal DNAs of the four transformants and the nontransgenic strain as the control were used as the templates for the PCR using the EGFP-specific primers. When the products were analyzed by the agarose gel electrophoresis, the expected 750bp amplified bands appeared. By fluorescence microscopy, distinct green fluorescence could be observed from the colonies of YLCs grown on RM agar plates and could not be observed from the colonies of YLCs which did not contain plasmid pCB-BEGFP. Expression of EGFP was detected by SDS-PAGE analysis of TCP of one transformants, as expected, a dark band of the protein was visualized at 27kD by Coomassie Brilliant Blue staining. These results indicated the EGFP gene had been integrated into the genome of transgenic Tremella fuciformis strains and was expressed successfully by regulation of gpd promoter of Agaricus bisporus.

  Genetic Diversity and Relationship of Cherry Germplasm by ISSR Markers Ai Chengxiang , Zhang Lisi , Li Guotian , Wei Hairong , Fan Jing , Liu Qingzhong 2008, 24 (4):  47-51.  Abstract + ( 1849 )   PDF ( 541821KB )   ( 1190 ) The genetic diversity and genetic relationships among 34 cherry accessions, including 3 interspecific progenies, 1 Prunus pseudocerasus, and 30 Prunus avium, were evaluated by ISSR markers. In ISSR analysis, a total of 149 bands were detected, among which 143 bands were polymorphic. The number of bands from each ISSR marker ranged from 6 to 12, with an average of 8.76. The PPB, PIC and Ne were 95.97%, 0.90 and 1.914, respectively. The ISSR-derived genetic similarity (ISSR-GS) ranged from 0.44 to 0.91, with a mean of 0.73. The cluster analysis indicated that all the 34 sweet cherry germplasm could be distinguished by ISSR markers. The genetic relationship cherry accessions were analyzed and 34 germplasm were classified on molecular level by clustered analysis, which provided reference for research in cherry germplasm.

  Molecular Marker for the Waxy Protein Subunits in Synthetic Hexaploid Wheat-Derived Populations Yang Songjie ，Yang Wuyun 2008, 24 (4):  52-57.  Abstract + ( 1910 )   PDF ( 599577KB )   ( 892 ) Synthetic hexaploid wheats, obtained by crossing tetraploid wheats (AABB) with Aegilops tauschii Coss (DD), containing genetic variability different from bread wheat, now have been widely used for genetic improvement of bread wheat. A total of 113 BC2F2:6 advanced lines derived from four CIMMYT synthetic hexaploid wheats, Syn768, Syn769 and Syn780, were used to detect Waxy protein by specific-primer PCR amplification and modified PAGE. The results showed that 8 of these cultivars and lines were the null Wx-B1 type and null frequency was 6.6%, and what is more, there was no other null types existing in the materials detected. The null frequency was different between the population from the three parents of synthetic hexaploid wheats. From this study, it could be concluded that the study on Waxy genotype of progeny from crosses of synthetic hexaploid wheat and bread wheat can provide useful information for improving bread wheat and be a useful tools in marker-assisted selection of wheat breeding programme.

  Genetic Relationships of Qianban Lotus Cultivars by RAPD Markers Guo Hongbo, Ke Weidong 2008, 24 (4):  66-68.  Abstract + ( 1712 )   PDF ( 390040KB )   ( 1241 ) Qianban Lotus, is a special type of lotus in Nelumbo nucifera Gaertn., which stamen and pistil are completely transformed petals and more than one-thousand petals may be observed during all the period of flowering. To discover the genetic relationships among current cultivars, 5 cultivars from three regions in Yunnan province and one from Yuquan Temple, Dangyang, Hubei province were investigated by employing randomly amplified polymorphic DNA (RAPD) markers. The selected sixteen primers amplified 141 bands in total, of which 81 were polymorphic (57.45%). Our result also exhibited that these cultivars had obvious genetic differentiation with relatively distant genetic distance. There is relatively high level of genetic diversity among these cultivars. No significant genetic differentiation was observed between those cultivars from Yunnan and the one from Yuquan Temple. The cultivars from Yunnan province were found to have obvious genetic differentiation, and the Yaoan lotus displayed its unique genetic characteristics at the most.

The results showed that good quality DNA would be obtained by modified SDS method and the 0.5% is better.

  Cloning of the Maize Starch Branching Enzyme SBEⅡb Gene and Constructing of its Over-expression Vector Fan Yali ,, Ruan Ying ,Li Jin ., Du Peifen, , Yao Yuanting, , Liu Chunlin 2008, 24 (4):  72-75.  Abstract + ( 2081 )   PDF ( 491119KB )   ( 1104 ) 【OBJECTIVE】In this article we cloned the Maize Starch Branching Enzyme SBEⅡb Gene and constructed its over-expression vector successfully. 【METHOD】The total RNA extracted from Waxy corn endosperm was as template, the Coding Regions of cDNA of the Maize Starch Branching Enzyme SBEⅡb gene was amplified Utilizing the method of RT-PCR.【RESULTS】The gene was successfully amplified. Homologous Analysis result by BLAST showed that the cloned fragment has 99% identity to the sequence of the gene in GenBank. The Coding Regions of SBEⅡb gene cDNA was 2719bp in length and encode 799 amino acids. 【CONCLUSION】The cloned cDNA was ligated into the plant express-vector pCAMBIA1303 . We successfully constructed a gene over-expression vector pCASBEⅡb through GUS activity test.

The factors affecting the inducibe culture and proliferate mediums and the methods of controlling contamination in culture were studied.The result indicated the suitable timing of taking shoot tips for culture were October to December, the most suitable inducibe medium was 1.0 mg/L 6-BA+0.25 mg/L NAA+1.0 mg/L GA3, the best proliferate medium was MS +4.0 mg/L 6-BA + 0.25 mg/L NAA. the best medium state is liquid-solid state, that is the uper layer’s component is MS + 1.0 mg/L 6-BA + 0.25 mg/L NAA + 1.0 mg/L GA3with 5 mm thickness and the under layer ’s component is MS + 1.0 mg/L 6-BA + 0.25 mg/L NAA + 1.0 mg/L GA3with 1cm thickness.the method of controlling contamination in culture was 75% alcohol 1min and 0.1% HgCl2 10min.

In this study, 19 out of 30 pairs of CAPS primers and 7 out of 10 SSR primers were selected, which gave stable and polymorphic amplification profiles. This two molecular techniques produced 98 bands, the results showed that the detectability of SSR is better than CAPS, 16.3 percent were form L.hirsutum, 10 percent were from L.pimpinellifolium, this two wild tomatoes could be used in genetics and breeding of tomato availably.

And the effect of different sterilized time, kinds of hormone and its combination on advenitious buds proliferation, plant regeneration and root regeneration were studied the in this paper. The results showed that explant sterilized in 0.1% aqueous mercuric chloride for 5 min had better effect, and the contamination rate was 31%, the germinating rate was 79%, inducement rate was 65.2%. MS+6-BA 2.0 mg/L+NAA 0.2 mg/L was suitable for advenitious buds proliferation from the leaf. The optimum medium was MS+6-BA 1.0mg/L+NAA 0.2mg/L. The best medium for rooting was 1/2MS+NAA 0.2mg/L. The comparatively perfect tissue culture regeneration system was formed.

Sequence analysis revealed that PCHS was 531 bp in total length and shared significant nucleotides identity 99% with GenBank databases, which contained four flower-specific expression regulatory elements. Intermediate vector pPCHS was constructed by inserting the promoter to the upstream of GUS in pB1121, where the original CaMV35S promoter was replaced by the PCHS promoter. The flower, stem, leaf ,and root of petunia hybrida were transformed via Agrobacterium tumefaciens. Histochemical localization of GUS activity indicated that the PCHS promoter could confer a flower-specific expression pattern to GUS.

The colned GAPDH gene (604 bp) was 95 %, 86 % and 84% identified with to M36650(Barley), AY109397 (maize), BT012693(tomato) , respectively. This partial sequence provided a good background on studying expression of many cereal genes.

In this study, a new method of CTAB was introduced which do best for characterization the purity and concentration of genome DNA. The result showed that it could effectively eliminate the affections of the secondary materials to extracted genome DNA from Okra, and which be suitable for RAPD analysis and other molecular maker.

The haematology and biochemistry indexes, organic histological maps of liver, kidney, intestines, lung, cerebra and testicle of parents and baby generation rats of the two groups were analyzed. The results showed that every index of the two groups did not have significant difference. It indicted that transgenic wheat xi’nong1376 with PSAG12-IPT gene did not have obviously bad influences to small white rats health and might be safety to human health.

  Isolation and Characterization of a Rice Gene OsRan1 Coding a Small G Protein Xi Jiang, Du Zhiru, Xu Yongju , Zhang Qi , Wan Jia, Jiang Bin, Xu Zhengjun, 2008, 24 (4):  109-114.  Abstract + ( 1709 )   PDF ( 660280KB )   ( 1235 ) The disadvantage circumstance is an important factor in impacting on the yield and quality of rice .The expression analysis of gene under stress helps a lot in studying of stress resistance of rice. A rice gene, OsRAN1, was isolated from rice treated with cold-stress by the fluorescence differential display (FDD) screening method，and its expression pattern was analyzed by simi-quantitative RT-PCR analysis under cold and salt stress. Its cDNA sequence (984bp) contains an ORF encoding a 221 amino acids protein (25.0KD)，and the protein belongs to Ran subfamily of the small G protein, for containing the typical GTPase structure. At the amino-acid sequence level, OsRAN1 exhibit high homology to TaRAN1 and AtRAN1 (98.19% and 92.76%). By simi-quantitative RT-PCR analysis, the expression of OsRAN1 at the transcription level was increased under the low temperature. Especially under high salt condition, the expression level in root shows a process of increase, and then decrease. We suggest the OsRAN1 gene could play an important role of stress response in rice.

  SRAP Analysis for the Genetic Diversity ofWatermelon Varieties Resources Zhang Aiping , Wang Xiaowu , Zhao Daqing 2008, 24 (4):  115-120.  Abstract + ( 2099 )   PDF ( 599113KB )   ( 1024 ) 【OBJECTIVE】Genetic relationships and classifications of a set of watermelon accessions were analyzed to provide the experimental support for utilizing effectively genetic materials for variety development.【METHOD】SRAP technique was adopted to analyze 64 watermelon accessions；【RESULTS】fifty-one primer combinations with clear band pattern and polymorphism were selected from over 700 primer combinations, These primer combinations produced 431 bands in which there were 243 polymorphic bands and the average frequency of the polymorphisms is 56.4%. When the wild-type material No.57 was removed, the frequency of the polymorphisms of other materials is 39.4%.The genetic similarity coefficient of the 64 accessions were calculated by the NTSYS software using 243 polymorphism SRAP bands. It’s ranged between 0.47 - 0.97. When excluding most variable four accessions including No.57, and range decreased to 0.87-0.97；【CONCLUSION】It showed that the watermelon has a limited genetic differentiation among the breeding lines and broaden genetic diversity is very important for watermelon breeding

  Comparative Analyses for Seed Dormancy QTL Between Maize and Wheat Lan Hai, Zhang Zhiming, Gao Shibin, Pan Guangtang, Rong Tingzhao 2008, 24 (4):  121-125.  Abstract + ( 1828 )   Through the comparisons of QTL for seed dormancy between maize and wheat, it was observed that there were remarkable conserved relationships of QTL for seed dormancy between maize and wheat. 7 QTL for seed dormancy which were detected on the chromosome 1, 3 and 8 of maize, are conserved with wheat. These results will be helpful to study the organization and evolution of maize and the grass genomes.

  Ampicillin on the Inhibition of Microbial Growth Determination Chen Yanfei 2008, 24 (4):  126-129.  Abstract + ( 1990 )   【OBJECTIVE】The antibiotic is a kind of secondary metabolite which is produced by the certain microorganisms in the process of growth.It has the ability that can suppress or kill some other microorganisms. The effect of ampicillin to different bacteria and different concentration of antibiotic to the same bacteria can be determined according to tength of the Bacteriostasis belt, and preliminary judge its antibacterial spectrum . 【METHOD】Using the law of filters the paper to mensurate the antibacterial spectrum of ampicillin.Colon bacillus ,staphylococcus aureus,bacillus are used.Along with ampicillin density increased.【RESULTS】 the longth of bacteriostasis belt grows gradually,and the effection of bacteriostasis become more strong.But the difference of Suppression effect that the same ampicillin to colon bacillus staphylococcus aureus,bacillus is not very certion.Throng restrain and transfers the peptide.【CONCLUSION】 ampicillin make bacteria die with cell wall disintegration.

  Cloning and Expression Analysis of a Ribosomal Protein from Rice（OsRPL14） Du Zhiru, Xi Jiang, Wan Jia, Jiang Bin, Xu Yongju, Zhang Qi, Xu Zhengjun, 2008, 24 (4):  130-134.  Abstract + ( 1647 )   Using a fluorescent differential display method, a gene was isolated from rice seedling. The full length cDNA was 565 bp and encoded a 134 amino acid(15.4kD). The results of similar analysis indicated that OsRPL14 had 82.84%-88.06% identities with other ribosomal proteins, named OsRPL14. The level of the gene expression was enhanced in response to cold(8℃)and dry. We suggest the gene might play important roles in stress respones.

  Determination of the Isomers of Vitamin E by Normal Phase High Performance Liquid Chromatography Xu Xiang, , Liu Guangmin, Gao Yanxiang 2008, 24 (4):  135-138.  Abstract + ( 2084 )   Tocopherols and tocotrienols were separated and determined by NP-HPLC to establish a simple and accurate method to determine the contents of the vitamin E isomers. The sample was purified and then separated on the Zorbax NH2 column. The mobile phase consisted of hexane and isopropanol. The detection was at 292nm. Seven isomers of vitamin E：α-tocopherol、α-tocotrienol、β-tocopherol、γ-tocopherol、γ-tocotrienol、δ-tocopherol and δ-tocotrienol were identified and calculated. The results showed that the corelation coefficients were from 0.9993 to 0.9998. The method suggested the high degree of precision, stability and the repeatability, and the recoveries were from 97.0% to 102.1%. This method was simple, accurate and suitable for the analysis of vitamin E in the whole berry oil of sea buckthorn.

  Study on Microwave-assisted Extraction of Natural Red Pigment from Pericarp of Pitaya Li Changhui, Yu Xiandong, Wan Xiaorong, He Shenggen, Huang Pengcheng, Zhang Weifeng 2008, 24 (4):  139-143.  Abstract + ( 2327 )   Microwave-assisted extraction（MAE）method was studied for the extraction of a natural red pigment from the pericarp of Pitaya (Hylocereus undatus Britt & Rose). Several factors, such as microwave power, extraction time and the ratio of solid to liquid were investigated, and the optimal MAE conditions were obtained from each factor and their orthogonal test. The MAE method was compared with the traditional solvent extraction for its advantages. The results showed that the optimum conditions for MAE is extracting for 80 seconds in the ratio of solid to liquid 1:3（g:ml）by microwave of 480 W with purified water as solvent. The MAE method is a fast, efficient, energy saving method, which is recommendable to the application for the extraction of natural red pigment from the pericarp of Pitaya.

  Nutrition Quality Analysis of Different Hawthorn Cultivars Bai Suhua, Zhu Yueying 2008, 24 (4):  144-146.  Abstract + ( 1830 )   To screen hawthorn cultivars which meet requirements for different nourishment and provide references for hawthorn breeding, the nutrition quality of 11 hawthorn cultivars were evaluated by measuring the nutritious parameters including total acidity, soluble protein, soluble sugar, and vitamin C. The fruits of different hawthorn cultivars contained significantly different nutrient contents, ranging from 1.625%~3.061% in total acidity, 5.165~8.714mg/g in soluble protein, 6.610~28.110% in soluble sugar, 55.87~139.70 mg/g in vitamin C. Among the cultivars evaluated here, Yanranghong exhibited excellent nutrient quality with highest evaluation index of 13 and Changkou reverse with poor score of 7. The significant difference among these hawthorn cultivars indicated that there is great potential to improve the nutritious quality of howthorn by breeding.
