Effects of Different Temperatures on POD Activities and Isozymatic Patterns during Early Somatic Embryogenesis in Longan

Abstract: The LC2 embryogenic cell line of the cultivar Honghezi of longan was used as materials for obtaining the synchronized embryogenic cultures of early somatic embryogenesis at different developmental stages which were treated with different temperatures (20 ℃ and 30 ℃,compared with 25 ℃) for analyses of the peroxidase (POD) activities and isozymatic patterns during longan early somatic embryogenesis. The results showed that with the differentiation of embryogenic callus,all the POD activities of the embryogenic cultures treated with three different temperatures decreased, and the POD activitives of the embryogenic cultures treated with high ( 30 ℃) or relatively low (20 ℃) temperatures at the successive developmental stages during early somatic embryogenesis showed higher than the comparison (25 ℃); the treatment of the temperature also had a great impact on the POD isozymatic patterns with the performance of the new bands for the emergence, which might be concluded that the changes of POD activities and isozymatic patterns in a law played a key role in the differentiation of early somatic embryogenesis, and keeping the normal development of somatic embryogenesis in longan.
