Microanalysis and Fast Analysis on Fatty Acids of Peanut Seed by Gas Chromatography

Abstract:

Abstract: The content of fatty acids is an important index for oil crops. The traditional methods of detecting fatty acids need more samples and more time, which is difficult to select hundreds of materials for breeders. To detect the fatty acids or to propagate is ambivalent when the material detected has few seeds. In this experiment, a fast microanalysis was studied for detecting fatty acids of peanut. A part of about 0.020g cut from one seed of peanut was used, extracted oil using the mixture (benzene: petroleum ether=1:1) for 5 minutes, and methyl esterificated using sodium hydroxide-methanol solution (c= 0.5 mol L-1) for 10 minutes, then the supernatant fluid was injected into the GC system. Using this method, the work which usually need several hours, now can be completed in 15 minutes, thus lots of time can be saved. By comparing this method with traditional method (GB/T 5009.6-2003, GB/T 17376-1998) for detecting fatty acid compositions, it was confirmed that the results got from this method was exact and credible.
