Influencing factors on microspore culture of Brassica napus L

Abstract: Thirty Brassica napus genotypes in Sichuan Chengdu were grown under uncontrolled temperature and light conditions, and their isolated microspore from anthers were used as explants in vitro culture. The influencing factors of microspore culture were studied and the main results showed that the temperature on the stage in microspore development 、disinfection method to bud 、and donor plant age are important influencing factors on microspore culture. Temperature was one of the best important influencing factors ,and the best temperature was 10-15℃,the highest embryo yield of the microspore developed on 10-15℃ was 300 per bud. The best disinfectant was Hgcl2 and the density was 0.1%.The best density of spore was 3-4 bud/dish .The donor plant age had obvious effect on embryo yield. Rate of embryo induction was increased according the increase of the donor plant age from 125 days to 150 days, and the rate of embryo induction was the highest 150 days in 2009. Then descend according the increase of the donor plant age.
