Abstract:

Abstract：Qingyuan partridge chicken is a light-body type breed with good meat quality, in order to explore the molecular mechanisms of its good meat quality, we chose CAPN1 gene as candidate gene for chicken meat tenderness, and Recessive White chickens as comparison in the present study. PCR-SSCP and sequencing methods were used to investigate single nucleotide polymorphisms in CAPN1 gene. Six genotypes were detected, i.e. AA, AB, AC, BB, BC and CC. Sequencing results revealed that AA genotype was coincidence with the sequence accessed in Genbank, there is a 5 bp deletion mutation at position 2418, a T/C point mutation at position 2446, a C/T mutation at position 2484, and a C/T mutation at position 2508, there is a 5 bp deletion mutation at position 2418, a T/C point mutation at position 2446, and a C/T mutation at position 2484. Allele frequencies and genotype frequencies distribution were significant different in the two chickens.
