Synthesis of Terbutaline artificial antigen and development of its mice polyclonal antiserum

Abstract:

The goal of this study was to synthesize artificial antigen of TBLaterol and obtain its mice polyclonal antiserum. Artificial antigen BSA-TBL and OVA-TBL were synthesized using butane-1,4-diol diglycidyl ether and EDC by linking carrier proteins BSA and OVA to TBL. The antigen BSA-TBL and OVA-TBL was identified by ultraviolet scanning, SDS-PAGE. The mice polyclonal antiserum was identified by ELISA and blocking ELISA. The results showed that the hapten TBL was successfully linked to carrier proteins. The animal immunization results showed that six BALB/c mice indirect ELISA titer against TBL were above 1×10-4 and the IC50 of No. 1 mice was 55.88 ng/mL . The high-titer mice polyclonal antiserum has been generated. Artificial antigen BSA-TBL and OVA-TBL were synthesized and sensitivity TBL polyclonal antiserum has been generated in this study. The artificial antigen of TBL and its mice polyclonal antiserum laid a foundation base for product the monoclonal antibodies and rapid test reagent against TBL.
