Abstract:

Abstract: to celeritily and nicetily identify the male sterility germplasms of ramie, The pollen vitality of 15 germplasm which were collected by our lab were examined by methods of dyeing and germination. The results showed that there are biggish difference in pollen vitality among different ramie germplams used different methods of dyeing. The effect of dyeing was not vary good by I2-KI, TTC and Aceto-Carmine. The pollen tube could be clearly seen used method of germination. So the best determine method of ramie pollen vitality was germination. When there was 10% saccharose concentration in the culture medium, 11 germplams could be grew pollen tube; When there was 20% saccharose concentration in the culture medium, 12 germplams could be grew pollen tube; When there was 30% saccharose concentration in the culture medium, all of 15 germplasms could not be germination. So the method of germination need 10%-20% saccharose concentration.
