Study on Culture in Vitro and Reversion for Vitrification on Seeds of Handeliodendron bodinieri (Lévl.) Rehd.

Abstract:

The method of culture in vitro was applied to study rapid propagation of Handeliodendron bodinieri (Lévl.) Rehd. which is an endangered wildlife catalogued as a national protected plant of China (Category I). Seeds of H. bodinieri were used as explants to induce sprouts and buds on medium contained different growth regulators. The results indicated that with suitable growth regulators, if seeds of H. bodinieri were collected and cultured in time germination rate could reach 96.7%. But with storage duration, the germination rate cut down gradually and wouldn’t have the ability of germination after half a year. The BA could make plants stronger and NAA could make plants taller. Buds grew well and the proliferation multiple was 3.43 on the medium of MS+BA1.0 mg/L+NAA0.2 mg/L. But with subcultures increasing, vitrification emerged. Vitrification could be reversed efficiently with changing MS medium into B5 medium and BA-NAA group into KT-IBA group. Orthogonal experiments results showed that the proliferation multiple and reversion for vitrification of buds were determined by the content of KT and medium was the second factor. The compounding medium of 0.75B5+KT1.0 mg/L+IBA0.05 mg/L+Sucrose 40000 mg/L could induce buds with high proliferation multiple and reversion of vitrification.
