Cloning and Sequence Analysis of the Complete Genome of Porcine Circovirus Type 1 Tianjin Strain

Abstract: Abstract: 【OBJECTIVE】To isolate and clone one porcine circovirus type 1 (PCV1) strain from Tianjin, and analyze its genome sequence. 【METHOD】A pair of primers for porcine circovirus type 1 (PCV1) was designed according to its sequences available in GenBank, the genome sequence of one PCV1 isolate was amplified by PCR assay with the tissues of the clinical diseased from a pig farm in Tianjin. Then, the complete genome of the PCV1 isolate was cloned, sequenced and analyzed. 【RESULTS】The genome sequence results showed that this genome is 1759 nt in size, and shared more than 98.2% identity of nucleotide with other PCV1 isolates selected from the GenBank. The sequences of the main open reading frame (ORF) revealed that there were about 97.5%~99.8% and 92.7%~99.9% identity of nucleotide in the ORF1 and ORF2 among the selected PCV1 isolates respectively. 【CONCLUSION】The present study indicated that there was a geographical correlation among the PCV1 isolates, although the nucleotide identity of the genomes and main ORFs of the selected PCV1 isolates was relatively high.
