The Connection between Concentration of total Polyphenol, Activity of Polyphenol Oxidase and Browning of Explants in Canarium album (Lour.) Rauesch

Abstract:

Abstract: The various tissues of Canarium album, (including old leavesmature leaves, young leaves, petiole relevantly, young shoots, flayed young shoots and flayed embryo), were collected in different month to detect the concentration of total polyphenol and the activity of polyphenol oxidase(PPO), then to reveal the relationship between the results and the browning of explants. We found that total polyphenol’s concentration and PPO activity in March were lower than that in other months; especially flayed young shoots had the lowest total polyphenol’s concentration and PPO activity, as well as flayed young shoots had the lowest browning rate which was helpful to the of Canarium album. The results revealed that total polyphenol’s concentration and PPO activity might be the critical factors to induce the browning of explants, so decreasing of them could enhance the efficiency of tissue culture.
