Abstract:

In order to find the appropriate determination methods of pollen viability of daffodil, three varieties of Narcissus spp．(‘Albany ’,‘Mount Hood’and‘Geranium’) of fresh pollen was treated with TTC staining, carmine acetate staining, in vitro germination to determine their viability. The results showed that: TTC staining is not suitable for Narcissus spp,, carmine acetate staining and in vitro germination could measure pollen viability of Narcissus spp．, between two methods carmine acetate staining can determinate the pollen viability of Narcissus spp. quickly, and germination in vitro is suit for accurate determination of pollen viability of Narcissus spp. .Compared between nine combinations of medium, the best medium combination is: 0.1g/ml sucrose +0.1×10-3g/ml boric acid.
