Effects of Different Phytohormones on Leaf Callus Induction of Phellodendron amurense Rupr.

Abstract:

The axenic leaf of Phellodendron amurense was used as explants to study the effect of different combinations of plant hormones on callus induction in P. amurense. The results showed that the callus induction could not be succeeded with 6-BA only. The induction rate was low when sole NAA or 2, 4-D was used, and the highest rates of the callus induction were 53.33% and 73.33%, respectively, with poor callus quality. When auxin and cytokinin were applied together, the callus induction was improved, and the optimum phytohormone combination was BA 1.5 mg/L+ 2,4-D 2.0 mg/L, which induction rate was 100%, and the callus grew luxuriantly.
