Abstract:

【OBJECTIVE】In order to find the more simple method of preparition for the layer feeder cells ;【METHOD】The blastocytes of 3.5 days embryo of KunMing mouse cultured on bone marrow mesenchymal stem cells were used to isolate and culture the KunMing ES cells. The colonies of ES cells were harvested to passage.【RESULTS】The growth of stem cells were identified was observed，and their biological vitality was determined by alkaline phosphatase activity assay and cell differentiation examination. 【CONCLUSION】The blastocytes of 3.5-dayembnyo from KM mouse can be cultured on the mouse bone marrow mesenchymal stem cells to develop into ES cells with maintenance of undifferentiated state in vitro.
