    Development of GC-MS Method for Detecting Clenbuterol Residues and Confirmation of Self-made Strip Performance

    GC-MS method of detection for Clenbuterol Hydrochloride (CL) residues in swine urine was established and compared with the method of self-made strip (CL-Strip). The results showed that the detection limits of GC-MS and CL-Strip were 0.5ng/ml and 1.0ng/ml respectively. The detection results of CL added in samples were consistent completely. The detection results of 100 negative swine urine samples and 18 positive swine urine samples by CL-Strip method were agreed with GC/MS method analysis results. It can be concluded that the sensitivity, accuracy and stability of CL-Strip method were about at the same degree as GC-MS method, and CL-Strip method was recommended to be extended for its advantages in convenience, direct-viewing, rapidity, sensitivity, specificity and accuracy.
    Optimization of Western Blot Condition for Detection of Protein Expression in Rabbit Artery

    Western blot is the most important method on studying and analyzing protein. Because it contains many steps, high background, unspecific bands, streaking and so on are the common problems in Western blot, which will influence the subsequent detection and quantitation of protein. To establish the best experiment condition for detection of protein expression in rabbit artery, VCAM-1(vascular cell adhension molecule-1) was detected. A rapid, specific and sensitive procedure has been successfully established, through optimizing several conditions which include SDS-PAGE, antibody dilution, temperature and time for antibody incubation, detection methods and so on. Also, three kinds of detection methods were used which contain DAB, enhanced chemilluminescence (ECL) with exposure captured on X-ray film or with cooled charged-coupled device (CCD) cameras. The sensitivity of three methods was compared. The results indicated that ECL has sensitivity 2-10 times that of DAB, and CCD cameras have 4 times sensitivity of X-ray and a more precise procedure. So, ECL-CCD cameras is a promising detection method of Western blot.

    The Use of Eugenol as an Anaesthetic for the Australian Longfinned Eel, Anguilla reinhardtii (Steindachner)

    Anaesthesia is important in handling large river eels during procedures such as measuring, tagging and surgery. Therefore, the use of eugenol as an anaesthetic for the Australian longfinned eel, Anguilla reinhardtii (Steindachner) was investigated with a range of concentrations (10-120 mg/L). 20-120 mg/L of eugenol was found to be safe and effective (P <0.05), which could induce anaesthesia in less than 10 min, and recover after anaesthesia also in less than 10 min with a recovery rate of 100%. The difference of induction times and recovery times among 40, 60, 80 and 100 mg/L was not statistically significant, but with and among other groups was significant (P <0.05). Interestingly, average induction times did not always drop as the concentration increased. The induction time decreased from 312.88 s to 252.25 s when the concentration increased from 20 mg/L to 30 mg/L, while increased greatly to 582.00 s when attained 40 mg/L. Between 40-120 mg/L, induction times were inversely correlated with concentrations (r =-0.84, P <0.05), and dropped again in general along with the increase of concentration. Mean recovery time was the shortest for 20 and 30 mg/L, and shorter for 120 mg/L, and showed correlations of a certain degree both with anaesthetic concentration and induction time (r = 0.52, P <0.05). In conclusion, eugenol is a safe and effective anaesthetic for the Australian longfinned eel. 20-30 mg/L of anaesthetic concentration which can induce anaesthesia in 4-5 min in average and recover after anaesthesia in 4-7 min is recommended.
    Isolation and Identification of Nitrite-oxidizing Bacteria

    Objective to study the isolation and characterization of the nitrite-oxidizing bacteria and identification by 16S rDNA. Methods one pure culture N3, which was enriched and isolated based on the procedures for nitrite oxidizer from sludge of a lake in Huzhou. Total DNA of pure culture was extracted and specific primers for the 16S rDNA of nitrite oxidizer were used for polymerase chain reaction (PCR). The amplified product was analyzed by agarose gel e1ectrophoresis and Sanger sequencing. Homology analysis was made by searching in Genbank with NCBI-Blast. Results the amplified fragment was 1441 bp for N3. 100% of N3 nucleotides were identica1 with the conserved fragment of Cellulosimicrobium cellulans (AY665978.1). Conclusion N3 phy1ogenetica1ly belonged to Cellulosimicrobium cellulans.

    Comparative study on winter environmental indices of beef cattle farms in Zhaosu and Baicheng counties in Xinjiang

    In order to compare environment changes of beef cattle farms in southern and northern Xinjiang in winter, environmental indices including the illumination, temperature, humidity, noise, concentrations of CO2, NH3, CH4, PM10 and TSP were measured in two typical beef cattle farms which were located in Baicheng County (BC) in southern Xinjiang and Zhaosu County (ZC) in northern Xinjiang. The indices were measured at 08:00, 12:00, 18:00 and 21:00 in 5 consecutive days. And the 24-hour change of the above indices was also measured in 3 consecutive days. Then these indices were compared between the two beef cattle farms. The results showed that the average illumination and temperature in BC farm, the average temperature and humidity in ZC farm were beyond the limit of the national standard of the Environmental Quality Standards of Livestock and Poultry Farms (NY/T 388—1999). In addition, both the average temperature and humidity in BC farm were extremely lower than that in ZC (P<0.01). Concentration of CO2 in ZC farm was far beyond the limit of NY/T 388—1999. In addition, concentrations of CO2 and NH3 were extremely lower in BC farm than that in ZC farm (P<0.01). The 24-hour change of the above parameters showed that the temperature in both the two farms was lower than the limit of NY/T 388—1999. The humidity in BC farm ranged from 52.7% to 86.4%, and the humidity of 12 hours in one day was over the limit of NY/T 388—1999; the humidity in ZC farm ranged from 69.4% to 97.4%, and the humidity of 18 hours in one day was over the limit of NY/T 388—1999. The concentration of CO2 in BC farm ranged from 687.6 to 1799 mg/m3 and it was over the limit of NY/T 388—1999 from 18:00 to 19:00; while the concentration of CO2 in ZC farm was between 1459.6~3392.0 mg/m3 and it was all over the limit of NY/T 388—1999 except for that at 05:00. All of the above results indicated that there were obvious differences of the environmental indices in the beef cattle farms between BC and ZC, the illumination and temperature in BC farm should be improved whereas the contradiction of increasing the temperature and decreasing the humidity and concentration of CO2 in ZC farm should be solved.

    In order to provide the guidance and reference on protection, development and utilization of genetic resources to the local rare bovine species in China, the experience of conservation and utilization on Mishima Cattle in Japan was introduced. The formation process, production and reproductive performances in Mishima Cattle were systematically stated via visiting Yamaguchi University and the Technological Center of Agriculture and Forestry, Yamaguchi Prefecture in Japan, and referring the related data of Mishima Cattle. Mishima Cattle were designated as Japanese‘national natural treasure’in 1928, which was regarded as the oldest Japanese Cattle and the most important bovine resource in Japan. Mishima Cattle had the ability of topquality beef with‘flake meat’. Mishima bull (and/or frozen semen) crossed Holstein cows to produce the hybrids with high yield and high-quality beef. However, Mishima Cattle not only matured later, possessed small body size, low reproduction rate, but also had smaller population size and high inbreeding coefficient. It is practical that Mishima bull and/or frozen semen cross beef and dairy breeds to produce high-quality beef.
    Review on Classification and Representative Species of Hemolysin

    【OBJECTIVE】Hemolysin as a Protein toxins, are prominent virulence factors of many pathogenic bacteria.But we have seldom found the reports about it in China; 【METHOD】So writer summarized some literature more than twenty years; 【RESULTS】name,classification and family of hemolysin have been written, especially three of representative species: α-toxin, Streptolysin O and HlyA; 【CONCLUSION】Nowaday, A relationship between the hemolysin and pathopoiesis of bacterium is obvious. For this reason, a distinct way may be found to treat the disease cause by bacterium, not to use antibiotic.
    Research of Cowside Tests for the Detection of Subclinical Ketosis in Cows during Early Lactation Period

    Ketosis is a common nutrition metabolic disease of dairy cows. Using ketone powder, urine analysis strip, milk analysis strip, kit detected the ketone concentration in milk and serum of dairy cows respectively, serum β-hydroxybutyrate 1200μmol/L as the threshold for normal cows and subclinical ketosis. Urine analysis strip and milk analysis strip diagnosis standard were 200μmol/L and 5mg/dl respectively. Serum β-hydroxybutyrate and glucose concentration of healthy cows were 726±43μmol/L, 3.42±0.05mmol/L respectively, subclinical ketosis cows were 2563±238μmol/L, 2.78±0.07mmol/L, difference was significant(P<0.05). Sensitivity and specificity of milk analysis strip were 67% and 92%, urine analysis strip were 57% and 85%,ketone powder were 60% and 94% respectively. It was an effectual method that cowside test diagnose the dairy cows subclinical ketosis with milk analysis strip.

    In order to supply shaping evidence of domestic animal nerves bear,the distribution of oxytocin on hypothalamus in 8~10 month old goat was determined by immunohistochemical SP method;The results demonstrated that oxytocin- immunoreactive(OT-IR)neurons were mainly distributed over nucleus paraventricularis and nucleus supra-optious; In addtion,OT-IR neurous was also found in nucleus perifornicals,nucleus ventromedialis, nucleus ventrolateralis, nucleus suprachiasmaticus, nucleus dorsmedialis, nucleus mamillaris and so on.The OT-IR cells were mainly spherical and ovoid , in addtion,some of them had projects.The positive cells which surrounded trabecula and located near the lumen were spindle-shaped. According to the all above describing, OT distribute on hypothalamus extensively.
    Morphology of Muskrat Scent Glands and Initial Culture of the Glands Epithelium

    The cystglands of matured muskrat in secreting or the cystglands induced secreting artificially were used. Based on the morphological observation, epithelial cells of the scent glands were isolated and cultured in vitro initially. The results showed that a great quantity of dispersed cells in single state was available for primary culture by digestion with 1mg/ml Collagenase IV for 5 h and 0.25mg/ml Trypsin for 15min at 37℃; DME/F12 containing 10% fetal bovine serum, penicillin(100U/ml)and streptomycin(100U/ml), as well as EGF at the concentration of 10ng/ml, could obviously improve the survival and proliferation of these cells; the epithelial cells could be serial subcultivated, freezed and thawed routinely as well; after the 5th passage, the epithelial cells were filled with many lipid dropleds, secreting and proliferating perfectly. These results suggest that it is feasible to produce artificial muskrat scent using cell engineering by isolating, culturing cystglanitd epithelial cell and furthmore establishing a musdrat cystgland epithelial cell line.
    Study on facture of transparent specimen in mice

    The results of making transparence specimen of mouse skeleton were compared respectively, making mouse death in 4 different methods and dyeing them in 3 methods. The results shows, drown or etherization to make mouse death, Alcohol-fixed, defatted with 3% potassium dichromate, vitrification with Potassium hydroxide, coloration of bone with Alizarin Red S, the glycerol-ammonia were respectively coloration of cartilage with Alcian blue, vitrification with glycerol which had different concentrations, were the best conditions for making transparent specimen. Temperature controlling could reduce the time of specimen-fixed.
    Impact on growth performance and fat deposition distribution of Lanzhou Fat-tailed sheep and Mongolian sheep with fat-tail removal

    In order to study growth performance and fat deposition distribution of fat-tail sheep with no tail, the impact on the growth performance and fat deposition distribution of ‘Lanzhou fat-tailed sheep’ and ‘Mongolian sheep’ with fat-tail removal was studied. 18 ‘Lanzhou fat-tailed sheep’ and 18 ‘Mongolian sheep’ which were healthy, at the same age and had almost the same weight were collected in the test. Nine ‘Lanzhou fat-tailed sheep’ and nine ‘Mongolian sheep’ were randomly selected to remove their fat-tail by different methods. ‘Lanzhou fat-tailed sheep’ used surgical method, and ‘Mongolian sheep’ used rubber ring ligation method. No tail sheep formed the treatment group and tail sheep made up the control group of each breed. The results showed that feed intake of each breed in treatment group was lesser than control group after slaughtered, while the average daily gain, live weight before slaughter and hot carcass weight in treatment group was greater than that of control group, but the difference was not significant(P>0.05). There was a highly significant difference in tail and rump fat weight of ‘Lanzhou fat-tailed sheep’ between control group and treatment group(P<0.01), and control group was highly greater than treatment group. The total body fat weight of ‘Lanzhou fat-tailed sheep’ in control group was also significantly greater than treatment group(P<0.05). The remaining parts of the fat weight of ‘Lanzhou fat-tailed sheep’ in control group were lesser than treatment group. Among them, there was no significant difference of the subcutaneous fat weight and perinephric fat weight between each group(P>0.05). The testicular fat weight, omental and mesenteric fat weight and internal fat weight showed significant difference between the two groups(P<0.05). There was also a highly significant difference in tail and rump fat weight of ‘Mongolian sheep’ between control group and treatment group(P<0.01), and control group was highly greater than treatment group. The subcutaneous fat weight, testicular fat weight, perinephric fat weight, omental and mesenteric fat weight, internal fat weight and total body fat weight of ‘Mongolian sheep’ showed no significant differences of the two groups(P>0.05). There was a trend in improving the average daily gain and decreasing feed intake of ‘Lanzhou fat-tailed sheep’ and ‘Mongolia sheep’ with tail removal. There was a little influence on fat deposition distribution with removing fat-tail of ‘Mongolian sheep’, while the fat are more deposited into the skin, testis, kidney and stomach and intestines with removing fat-tail of ‘Lanzhou fat-tailed sheep’. What’s more, the total body fat weight of ‘Lanzhou fat-tailed’ sheep was significantly declined.
    Effcets of Insulin-Like Growth Factor 1 on the Proliferation at Different Generation of Antler Organic Center Cells

    【Objective】To investigate the effects of insulin-like growth factor 1 (IGF1)on the proliferation of antler organic center cells from various passages in vitro at 60 days growing period of antler. 【Method】Antler organic center cells were isolated from sika deer antler of growing 60 day and cultured. The second, fifth and eighth population cells were incubated with 0, 1, 3, and 10 nM IGF1. Thymidine incorporation assay was applied after 24 hours of culture respectively to determine the disintegration per minute ( DPM) value.【Results】The proliferation of cells from growing 60 day’s antler in the experimental groupwas significantly higher than that of the control group (p<0.01). The average and maximal DPM value of the proliferation of cells in the experimental group was the highest in the second population cells (31918 and 39818 DPM/mg respectively), decreased in the fifth population cells materially (5455 and 6815 DPM/mg), and decreased in the eighth population cells again (4030 and 4838 DPM/mg). 【Conclusion】The results show that IGF1 can enhance the proliferation of antler organic center cells cultured in vitro. The sensitivity of IGF1 on the antler organic center cells from various passages at 60 days growing period of antler in vitro.

    It was the first time that microadditives were used in pig-manure composting. The results of additives on pig-manure composting indicated that these additives could not only accelerate the compost’s maturity but also improve its quality due to the rapid growth and reproduction of microbes in pig-manure caused by additives. Lanthanon guoguang was the best among them.
    Eukarytic Expression and Identification of Nucleocapsid Protein N Gene of Canine Distemper Virus

    According to the published sequence of N gene of CDV, a pair of primers was designed and synthesized, which can amplify CDV N gene. N gene’s cDNA fragment of about 1.6kb was obtained through RT-PCR from FOX TA strain of CDV, and was cloned into pIREShyg Vector Eukarytic Expression Vector named as pIRES-N was constructed. pIRES-N was transfected into CHO-K1 cells with coprecipitation, and positive cells were screened with hygromycine selection. The N gene protein was expressed in the CHO cells proved by IFA. The N gene was transcripted as proved with RT-PCR. CHO/CDV-N cell strain was constructed, which provided evidence for research of CDV serumal detection and gene vaccine.
    The Relation between the Virus-seed of Infectious Bronchitis Virus(M41)and its Immunogenicity

    Whether different generations of IBV M41 strain could be used as seed for producing vaccine was evaluated. In this article IBV M41 strain was handed down from 1 to 8 generation. The 3rd, 5th and 7th offsprings were chosen for evaluating their immunogenicity by detection of their EID50. Vaccines to the three generations were prepared and used for inoculating the SPF chicken. The results indicated that the three vaccines could protect the birds and the protection rate was all above 9/10. The controls were infected and showed signs of the disease. The antibody titers of them were assayed at post-inoculation and the results indicated that the mean antibody titer rose to 3 times of that of the pro-inoculation. Different generations of IBV M41 strain still kept good immunogenicity and could be used as product seed for preparing IB vaccines.
    The study on the Forelimb Bone of Alpaca(lama pacos)

    The forelimb bone of alpaca were studied by the method of gross anatomy, the comparative study with the forelimb of camel, equine, bovine, ovine, and swine were done at the same time. The result shows that the alpaca forelimb consists of scapula, humeri, 0ssa antebrachii(radius, ulna),0ssa Carpi,0s metacarpale,0ssa digiforum mamus. The composing and structure of alpaca are same to the bone of camel and similar with the bone of bovine, ovine, and swine in some degree. The alpaca scapula, and 0s metacarpale, 0ssa digiforum mamus were similar with the scapula of bovine. The humeri, 0ssa antebrachii(radius, ulna)were similar to the bone of equine. From above depicting and comparison, we concluded that the forelimb of alpaca were same with the forelimb of camel but smaller in form, were similar with the forelimb bone of equine, bovine, and ovine, were different to the bone of swine.
    Effects of melatonin on fur Quality and growth performance of Rex rabbit

    80 rex rabbit (45-50 days old) were randomly divided into 4 groups of 20 rabbit each. The levels of hypodermic embedmenting MLT were 2.33, 3.5, 7 and 0 milligrams per animal. The trial last for 80 days. The result showed that the weight gain, hair-follicle differentiation and fur density of the embedmented MLT rabbits were increased and the skin maturing was promoted significantly (p<0.01); The live weight, fur density and skin thickness of 130 days old rabbits embedmented 3.5~7 milligram MLT reached to that of 150 days old. In other words,the commercial rex rabbit embedmented MLT can reached to slaughter weight 20 days ahead of time.
    Extraction and Effects of Isatisroot Polysaccharide(IRPS) on the Porcine Parvovirus(PPV) in Vitro

    The experimental boil alcohol extraction isatisroot polysaccharide, and PK-15 cells cultured porcine parvovirus (PPV), by observing cytopathic effect (CPE) to evaluate isatisroot polysaccharide on the porcine parvovirus blocking role and inhibition used. The results showed indigowoad root polysaccharides in the cell concentration of security, porcine parvovirus obviously blocking role and the role of inhibition, the smallest concentration of 2.09μg/ml block, minimum inhibitory concentration for 4.19μg/ml.
    Effects of Bacteria and Accessories of Chicken Manure Compost

    In order to study the influence of bacteria and accessories of chicken manure compost, the experiment was conducted to study the changes of temperature,Escherichia coli numbers and the odor during chicken manure compost by adding sawdust, rice husk, yeast and lactic acid bacteria respectively. The results showed that: (1) The highest fermented temperature of yeast group reached 67.6℃, 6.9℃higher than in lactic acid bacteria group, and the fermented time lasted 16 days over 55℃, 5 days more than in lactic acid bacteria group, which better than in control group (55.9℃,5 days); (2) The highest fermented temperature in sawdust group 2.2℃higher than in rice husk group,and time lasted 3 days more than in rice husk group over 55℃; (3) The number of E. coli was reduced from 105 to 102 per gram in yeast group and lactic acid bacteria group. The odor was disappeared respectively in 7th and 8th day. The effects showed that yeast and sawdust were best for chicken manure compost in the experiment.
