摘要: 摘要:胶原酶消化法分离和培养家兔子宫内膜细胞,大肠杆菌细菌脂多糖(LPS)诱导子宫内膜细胞炎症,以培养细胞的形态学和传统炎症指标作为炎症发生和发展的判断标准。以不同浓度的LPS作用于细胞,收集不同时段培养上清液,ELISA法测定TNF-α、IL-1β的含量。结果表明,100ng/mL LPS是体外培养的子宫内膜细胞诱导炎症的最适浓度。炎症模型组培养上清液中TNF-α、IL-1β的含量比同期空白组高(P<0.01),表明炎症模型建立成功。

Abstract: Abstract: The primary cultured endometrial cells in rabbits were isolated and purified by collagenase digestion method, cultured in vitro, and induced inflammation by lipopolysaccharide (LPS). The morphology and traditional inflammation factors of the cells were measured as the development criteria of inflammation. The cells were treated with gradient concentrations of LPS, the culture supernatants were collected in different period, and the contents of TNF-α, IL-1 β were determined by ELISA. The results showed that 100 ng/mL LPS was the optimal concentration to induce the inflammation of cultured endometrial cells. The supernatant TNF-α, IL-1 β in the inflammatory model group were higher than those in control at the same period (P<0.01), the inflammatory model of the endometrial cells was created successfully.
