桑树同源多倍体基因组DNA甲基化状态的MSAP分析

DNA Methylation in Polyploidy Mulberry by MSAP Analysis

摘要: DNA甲基化被认为是研究同源多倍体表观遗传现象的最佳途径。以‘新一之濑’、‘陕桑305’和‘陕桑402’为材料,研究桑树同源多倍体基因组DNA甲基化水平及变化模式。共筛选出21对引物组合应用甲基化敏感扩增多态性技术(MSAP),‘新一之濑’、‘陕桑305’和‘陕桑402’分别检测到507、507和511个位点。结果表明:从总体上看,‘陕桑305’、‘陕桑402’的总甲基化水平与‘新一之濑’的差异不大,桑树多倍化后甲基化模式发生了大量调整,主要以去甲基化类型(B型)为主,其次是过或超甲基化类型(C型)。由此推测,桑树同源多倍体可能通过DNA甲基化,在基因组加倍后,重新调控基因表达,从而表现出优势农艺性状。

Abstract: DNA methylation was considered to be the best way to study on epigenetic phenomenon of autopolyploid. The levels and patterns of methylation in mulberry were studied based on ‘Shinichinose’, ’Shansang 305’and’Shansang 402’. 21 primers combinations were selected to apply MSAP analysis. 507, 507 and 511 sites were detected respectively in ‘Shinichinose’, ’Shansang 305’and’Shansang 402’. The results showed that the total methylation level had little differences in ‘Shinichinose’,’Shansang 305’and’Shansang 402’ as a whole. The methylation patterns of mulberry had a large amount of adjustment after polyploidization, type B was the most and type C following behind in all types. It is conjectured that the DNA methylation maked the genome double and regulated gene expression to appear the advantages of agronomic traits.
