几丁质酶基因chiB 的克隆及表达分析

Cloning and Expression of A Chitinase Gene chiB

摘要: 几丁质酶能够通过破坏昆虫组织中的几丁质起到抗虫作用。为获得高表达的植物几丁质酶基因载体,通过同源克隆的手段,从粘质沙雷氏菌中分离到了chiB 基因,长度为992 bp,构建了pET28 表达载体,并用IPTG诱导了该基因的表达。经分析发现该基因表达产物属于糖基水解酶(glycosyl hydrolases)18家族,并利用pCAMBIA2300构建了植物表达质粒pCAM-35S-chiB。

Abstract: Chitinase can play the role of insect resistance by destroying chitin in insect tissues. In order to get the plant expression plasmid of chitinase, homology cloning method was used to separate chiB gene from Srratia marcescens, the length was 992 bp; pET28 expression vector was constructed and the gene expression was induced by IPTG. The analysis showed that the gene expression product belonged to glycosyl hydrolases family 18, and plasmid pCAM-35S-chiB was constructed by pCAMBIA2300.
