赤芍对金黄色葡萄球菌增殖和细菌毒力因子表达的影响

Influence of Radix paeoniae Rubra Decoction on Secretion of Virulence Factors and Proliferation of Staphylococcus aureus

摘要: 为探讨赤芍水煎剂在体外对金黄色葡萄球菌增殖及细菌毒力因子(α-溶血素、肠毒素)分泌的影响,在体外以不同浓度的赤芍水煎剂作用于金黄色葡萄球菌,记录细菌的生长曲线;用Western-blot 方法检测经赤芍水煎液处理后的细菌培养液上清中细菌主要的毒力因子α-溶血素、肠毒素A、肠毒素B的含量。结果表明赤芍水煎剂在0~10 mg/mL 对金黄色葡萄球菌ATCC29213 和ATCC43300 菌株的增殖没有明显的影响,但是赤芍水煎剂在不抑制细菌增殖的浓度下,能够降低金葡菌ATCC29213 和ATCC43300 菌株毒力因子α-溶血素、肠毒素A、肠毒素B的分泌表达。本研究结果提示赤芍水煎液中具有抗细菌毒力因子的药物成分。

Abstract: To study the influence of Radix paeoniae Rubra decoction on the secretion of virulence factors (α- Hemolysin, HLα; staphylococcal enterotoxins) and the growth of Staphylococcus aureus in vitro, Staphylococcus aureus was treated with Radix paeoniae Rubra decoction at different concentrations. Then the bacterial growth curve was recorded. Western-blot was performed to investigate the content of HLα, staphylococcal enterotoxins A (SEA) and B (SEB) in the supernatant of bacterial culture solution. The results showed that Radix paeoniae Rubra decoction could not inhibit the proliferation of Staphylococcus aureus (ATCC29213 and ATCC43300) obviously when the concentration ranged from 0 to 10 mg/mL. However, it could decrease the secretion of HLα, SEA and SEB in a dose-dependent manner. The results indicated that there was a potential anti-virulence ingredient in Radix paeoniae Rubra decoction.
