正交设计优化北苍术组织培养研究

Tissue Culture of Atractylodes chinensis by Orthogonal Design

摘要: 为了拯救濒危植物——北苍术,保护种质资源,在山西省中条山和太行山采集北苍术,以北苍术的种子为材料获得无菌试管苗,分别利用根、胚轴、叶片为外植体,研究了外植体对愈伤组织诱导率的影响;应用正交试验法研究6-卡基腺嘌呤(6-BA)、激动素(KT)、萘乙酸(NAA)及其配比对北苍术无菌苗增殖的影响;同时研究了NAA浓度对北苍术生根的影响。结果表明:以胚轴为外植体在1/2MS 2,4-D 2.0 mg/L KT 1 mg/L 蔗糖3%的培养基中更有利于形成愈伤组织从而长成完整植株;北苍术的最适增殖培养基为:1/2MS 6-BA 2 mg/L KT 1 mg/L NAA 0.5 mg/L 蔗糖3%;最适生根培养基为:1/2MS NAA 0.5 mg/L 蔗糖3%。

Abstract: In order to protect the wild resources of Atractylodes chinensis and realize their artificial cultivation,taking the rootand hypocotyl of Atractylodes chinensis as explant,the author studied the callus inducement and differentiation, rooting of adventitious buds. Rooting and transplanting of test-tube seedlings of Atractylodes chinensis, finally established its tissue culture technique system. The results show that the optimum medium for callus inducement and differentiation is 1/2MS (Sugar content is 30 g/L) 6-BA 2 mg/L KT 1 mg/L NAA 0.5 mg/L, 1/2MS (Sugar content is 30 g/L) 6-BA 2 mg/L KT 1 mg/L NAA 0.5 mg/L GA 0.5 mg/L is the optimum medium for rooting.
