树脂纯化武当道茶多糖的研究

Polysaccharide Purification from Wudang Taoism Tea by Resin

摘要: 为探讨树脂在武当道茶多糖脱色和脱蛋白过程中的效果及DEAE-纤维素-52 层析柱分离武当道茶多糖的效应,以单因素实验为基础,对样品上样流速、温度和料液比进行L9(34)正交实验,采用蒽酮-硫酸法测定多糖含量、考马斯亮蓝法测定蛋白含量。树脂201x7 在料液比11:1、上样流速1.0 mL/min、50℃条件下,武当道茶多糖脱色率为87.49%,脱蛋白率为43.27%,多糖保留率为89.12%。树脂201x7 纯化后的武当道茶多糖经DEAE-纤维素-52 层析柱分离得到5 个不同组分。上述方案稳定性好效果明显,可用于武当道茶茶多糖的开发和利用。

Abstract: The paper aims to study the effects of resin on decolorization and deproteinization of Wudang Taoism tea, and the effect of DEAE-52 cellulose column chromatography on polysaccharide isolation from Wudang Taoism Tea. Based on single-factor experiments, orthogonal tests were employed to evaluate three factors including sample loading flow rate, temperature and ratio of material to water according to L9(34) scheme. The anthrone-sulfuric acid method and the coomassie brilliant blue method were eparately applied for the content detection of polysaccharide and protein. When the sample was absorbed by 201x7 in the condition of 50℃ , 1.0 mL/min and the ratio (material to water) of 11:1, the rates of decolorization, deproteinization and polysaccharide retention was 87.49% , 43.27% and 89.12% , respectively. Five polysaccharide fractions were isolated by subsequent DEAE-52 cellulose column chromatography. The scheme is steady and effective, and suitable for further development and utilization of olysaccharide from Wudang Taoism tea.
