SRAP标记对甜菜抗褐斑病品种的聚类分析

Cluster Analysis of Sugar Beet Resistance to Cercospora Leaf Spot with SRAP Makers

摘要: 为进一步鉴定分析抗褐斑病性较好的国内外甜菜品种,为日后筛选、繁育优良抗病品种提供依据,本研究采用田间调查与分子标记相结合的方法,利用筛选出的12 对SRAP引物组合对25 份国内外品种进行分子标记检测,通过SRAP-PCR扩增,共产生扩增带104 条,67 条多态性条带,平均多态性比为64.4%。遗传距离平均为0.4670,遗传相似系数平均为0.6269。田间调查结果显示:褐斑病级数为2 的品种有9个,褐斑病级数小于2的品种有3个。聚类分析结果显示:在遗传距离0.20处,可将25份供试材料分为3 个类群,其中第一类群又分为2 个亚类。聚类结果与田间调查结果相一致。结果表明,‘TY305’(国内)、‘TY309’(国内)、‘w5-1’(国内)抗褐斑病性较好,SRAP分子标记用于划分甜菜抗褐斑病性的品种有一定参考价值。

Abstract: The aims are to identify and analyze domestic and foreign sugar beet varieties with better resistance to Cercospora leaf spot, and provide a basis for screening and breeding excellent disease-resistant varieties in the future. Field investigation and molecular marker were used in this study. 12 pairs SRAP molecule markers were used to detect 25 domestic and foreign varieties. A total of 104 unambiguous bands were obtained by SRAP-PCR, 67 of which were polymorphic. The average ratio of polymorphic bands was 64.4%. The genetic distance was 0.4670, and the genetic similarity was 0.6269. The results of field investigation showed that nine varieties belonged to the level 2 Cercospora leaf spot disease, and three varieties had Cercospora leaf spot disease level lower than level 2. The cluster analysis showed that 25 genotypes were divided into three cluster groups based on cluster analysis (at intercept of 0.20), and the first group was divided into two sub-groups. The clustering results were consistent with the field survey results. The results showed that‘TY305’(domestic), ‘TY309’(domestic) and‘w5-1’(domestic) had better resistance to Cercospora leaf spot. SRAP molecular markers had reference value for dividing Cercospora leaf spot resistant varieties of sugar beet.
