RP-HPLC法测定柚皮中柚皮苷和橙皮苷的含量

RP-HPLC Determination of Naringin and Hesperidin Content in Pummelo Pericarp

摘要: 为优化柚皮中柚皮苷、橙皮苷的同时测定方法,比较不同来源柚皮中柚皮苷和橙皮苷的含量差异,采用Luna C18 (2) (4.60 mm×150 mm,5 μm,100 ?)为色谱柱,水:乙腈=81:19(以磷酸调pH 2.4)为流动相,流速为1.0 mL/min,柱温为30℃,检测波长为283 nm的高效液相色谱方法,对柚皮中的柚皮苷、橙皮苷含量进行测定。结果表明,该分析方法的柚皮苷和橙皮苷分别在0.0247~0.989、0.0282~1.130 μg 范围内具有良好的线性关系,r=0.9999;柚皮苷和橙皮苷的平均回收率分别为91.46%、92.66%,RSD 分别为1.81%、1.22%;经该法测定,不同来源柚皮中柚皮苷含量差异达极显著水平,但均未检测到橙皮苷。该方法简便、准确,可同时测定柚皮中柚皮苷和橙皮苷的含量。

Abstract: To optimize the simultaneous determination method of naringin and hesperidin, we compared the content difference of naringin and hesperidin of pomelo peel from different sources. Naringin and hesperidin content were determined by RP-HPLC on Luna C18 (2) column (4.60 mm×150 mm, 5 μm, 100 ?) at 30℃. The mobile phase was pure water-acetonitrile (81:19), adjusted to pH value of 2.4 with phosphoric acid. The UV detection wavelength was set at 283 nm. The flow rate was 1.0 mL/min. The calibration range of naringin and hesperidin was 0.0247- 0.989 μg (r=0.9999) and 0.0282- 1.130 μg (r=0.9999), respectively. The average recoveries of 3 levels of naringin and hesperidin were 91.46% and 92.66%, corresponding RSD were 1.81% and 1.22%. Through this method, the difference of naringin content of pomelo peel from different sources reached extremely significant level. But hesperidin was not detected. This method is simple and accurate, and can simultaneously determine naringin and hesperidin content in pomelo peel.
