不同年限唐古特瑞香不同药用部位活性物质初探

Preliminary Study on the Active Substances of Different Medicinal Parts of Daphne tangutica Maxim of Different Planting Years

摘要:

为对比不同地区、不同年限野生‘唐古特瑞香’根、茎中3种成分含量、变化规律,利用高效液相色谱仪(HPLC),应用ZORBAX SB-C18(4.6 mm ×150 mm)色谱柱,流动相使用乙腈︰0.05%磷酸(10:90)和甲醇︰水(20:80),流速为0.9 mL/min,检测波长为265 nm和327 nm,柱温为40℃,进样量为20 μL。建立的HPLC分析体系线性范围良好(R2>0.999),精密度、稳定性、重复性RSD均小于2%,加样回收率在95.10%~95.84%。含量结果分析表明,同一部位的样品,紫丁香苷的含量在第5年显著增加(P<0.05);互助地区根皮、茎皮和哈溪地区茎皮中祖师麻甲素、7-羟基香豆素,在第5年含量显著减少(P<0.05)。互助地区茎皮和哈溪地区根皮中祖师麻甲素与7-羟基香豆素含量次序分别为5年生>8年生>3年生,5年生>3年生>8年生(P<0.05)。同一年限的样品,除茎皮中祖师麻甲素、7-羟基香豆素和紫丁香苷含量显著高于根皮外(P<0.05),哈溪地区茎皮祖师麻甲素含量显著高于互助地区的根皮和茎皮含量(P<0.05);5年生和8年生祖师麻甲素与3年生7-羟基香豆素和紫丁香苷的含量依次为互助茎皮>哈溪茎皮>互助根皮>哈溪根皮(P<0.05);5年生和8年生紫丁香苷的含量依次为互助茎皮>互助根皮>哈溪茎皮>哈溪根皮(P<0.05)。‘唐古特瑞香’同一部位样品中多数成分含量在第5年较高;同一年份茎皮中多数成分含量高于根皮;哈溪地区5年生唐古特瑞香茎皮中成分含量最高。

Abstract:

This study is to analyze the contents of three components and their changes in the rhizomes and stems of wild Daphne tangutica Maxim grown in different regions and of different planting years. High performance liquid chromatography (HPLC) and ZORBAX SB-C18 (4.6 mm ×150 mm) chromatographic column were used. The mobile phases of acetonitrile: 0.05% phosphoric acid (10:90) and methanol: water (20:80) were adopted. The flow rate was 0.9 mL/min, and the detection wavelength was 265 nm and 327 nm, respectively. The temperature was 40℃, and the sample load was 20 μL. The established HPLC system had good linear range (R2 > 0.999), the relative standard deviation (RSD) of precision, stability and repeatability were all less than 2%, and the recovery rate was 95.10%-95.84%. The content analysis showed that the content of syringin in the same part increased significantly in the fifth year (P< 0.05); the content of Daphentin and 7-hydroxycoumarin in the root bark and stem bark in Huzhu area and the stem bark in Haxi area decreased significantly in the fifth year (P< 0.05). The order of Daphentin and 7-hydroxycoumarin content in stem bark in Huzhu area and root bark in Haxi area was 5-year-old > 8-year-old > 3-year-old and 5-year-old > 3-year-old > 8-year-old, respectively (P< 0.05). For the same year samples, except that the content of Daphentin, 7-hydroxycoumarin and syringin in stem bark were significantly higher than those in root bark (P< 0.05), the content of Daphentin in stem bark in Haxi area was significantly higher than that in root bark and stem bark in Huzhu area (P< 0.05). The content of Daphentin of 5-year-old and 8-year-old and the content of 7-hydroxycoumarin and syringin of 3-year-old were in the order of Huzhu stem bark > Haxi stem bark > Huzhu root bark > Haxi root bark (P< 0.05); the content of syringin of 5-year-old and 8-year-old were in the order of Huzhu stem bark > Huzhu root bark > Haxi stem bark > Haxi root bark (P< 0.05). The content of most components in the same part of Daphne tangutica Maxim were higher in the fifth year; and the content of most components in stem bark was higher than that in root bark in the same year plant; the content of components in the stem bark of 5-year-old Daphne tangutica Maxim in Haxi area was the highest.
