Effects of Different Chemicals on Nitrogenase Activity of Monochamus alternatus Hope

摘要:

目前,松褐天牛是国内外林业有害生物防治中的一个难题,基于松褐天牛为食木昆虫,具有自身固氮能力,通过筛选出对松褐天牛固氮活性具有较大影响的药剂,来破坏其固氮能力导致松褐天牛缺乏氮素营养而死亡,从而来开辟新的防治途径。本研究采用乙炔还原法及酶联免疫试剂盒测定了不同药剂处理下的松褐天牛肠道固氮菌和不同生长阶段(成虫、幼虫及卵)虫体的固氮酶活性差异,结果表明,阿维菌素和NH4Cl处理对松褐天牛成虫固氮酶活性的影响较大,在药剂处理下的固氮活性仅为0.1 IU/g和0.08 IU/g,与对照相比固氮酶活性分别减少1.82 IU/g和1.85 IU/g;而固氮活性最强的菌株EH在NH4Cl处理下的固氮活性仅为1.2 nmol/(mL·h),比正常情况下的固氮活性减少了50.9 nmol/(mL·h)。因此,理论上NH4Cl可应用于林间防治松褐天牛。

Abstract:

At present, Monochamus alternatus is a challenge in the prevention and control of forestry pests at home and abroad. Based on the fact that Monochamus alternatus is a wood-feeding insect and has its own nitrogen-fixing ability, the nitrogen fixation ability of Monochamus alternatus can be destroyed by screened chemicals, resulting in the death of Monochamus alternatus due to the lack of nitrogen nutrition. This is a new way of pest control. The authors used acetylene reduction method and enzyme-linked immunosorbent assay kit, to determine the difference of nitrogenase activity between intestinal nitrogenase bacteria of Monochamus alternatus and insects at different growth stages (adults, larvae and eggs) under different chemical treatments. The results showed that the treatment of abamectin and NH4Cl had great influence on nitrogenase activity of Monochamus alternatus adults. The nitrogenase activity under both the two chemical treatments was only 0.1 IU/g and 0.08 IU/g, respectively. Compared with that of the control, the nitrogenase activity was decreased by 1.82 IU/g and 1.85 IU/g, respectively. Strain EH had the strongest nitrogen fixation activity, but its nitrogen fixation activity was only 1.2 nmol/(mL·h) under NH4Cl treatment, 50.9 nmol/(mL·h) less than the normal nitrogen fixation activity. In theory, NH4Cl can be used to control Monochamus alternatus in forests.
