RAPD标记对25份柑桔资源及其芽变系的鉴定和多样性分析

Identification and Genetic Diversity Analysis of 25 Citrus Resources and Its Sports Strains by RAPD Technique

摘要: 利用随机扩增多态性DNA(RAPD)技术,对几份柑橘资源及其芽变系共计25份材料进行了鉴定和遗传多样性分析,从70个引物中筛选出多态性好的10个引物组成单引物和双引物对基因组进行扩增,共扩增出142条谱带,其中多态带72条,共同带70条,平均每个单引物出现多态带6.8条,每对双引物出现多态带1.3条。利用UPGMA法分析发现,这些基因型间的遗传相似系数在0.834~0.966之间,在此基础上,构建了各基因型的遗传关系树状图。结果表明RAPD标记可以准确鉴定出各柑橘资源及其芽变系,这25份柑橘资源代表了25个基因型。较高的遗传相似系数揭示了柑橘资源间狭窄的遗传背景。

Abstract: The genetic diversity and identification of citrus germplasm resources and its mutants was evaluated by Randomly Amplified Polymorphic DNA(RAPD)technique. Ten of seventy random primer and several mixed primers could produce 142 bands, 72 of which were polymorphic. 6.8 polymorphic bands were displayed by every single primer and 1.3 polymorphic bands by every mixed primer. Cluster analysis showed that similarity coefficient between 0.834 and 0.960 of these 25 genotypes by UPGMA(un-weighted pair group method with arithmetic)analysis. And based on those data, the dendrogram of these genotypes was constructed. The results revealed RAPD marker is suitable for identify sports from its original strains, 25 citrus resources indicate 25 genotypes. The high similarity coefficient showed that there is narrow genetic background among citrus resources in the experiment.
