ITS序列在苦荞麦种质资源鉴定中的应用

Application Research of ITS Sequences in Fagopyrum tataricum Identification

摘要:

为了更精确有效地鉴定苦荞麦种质资源,根据ITS序列通用引物,以苦荞麦基因组DNA为模板通过PCR方法扩增出ITS(internal translation spacer)序列,在克隆测序的基础上对荞麦属5个栽培苦荞麦中进行分子鉴定与亲缘关系分析。结果表明:(1)5个供试材料ITS序列的变异小,序列长度在584~590 bp之间,G+C含量高,为67.5%~68.5%。(2)通过聚类方法分析发现‘建德苦荞麦’和‘红花苦荞麦’聚为一支,‘西荞2号’和‘西荞3号’聚为一支。

Abstract:

In order to identify germplasm resources of Fagopyrum tataricum, in this study the ITS general primers of sequence was used, F. tataricum genomic DNA as a template was amplified by PCR out of ITS sequences (internal translation spacer), the cloned sequence was based on 5 cultivars of F. Tataricum for molecular identification and molecular analysis of genetic relationship. The results showed that: (1) five materials were a small variation of ITS sequences, the length of sequences between 584-590 bp, G+C content was high up to 67.5%-68.5%. (2) Using different clustering analysis showed that ‘Jiande’ F. Tataricum and ‘Honghua’ F. Tataricum, ‘Xi 2’ F. Tataricum and ‘Xi 3’ F. Tataricumt had a closed genetic relationship.
