石栗树茎段组织培养的研究

摘要:

建立石栗组织培养的技术体系,筛选出适合诱导愈伤组织和愈伤组织分化的培养基。从外植体消毒,激素配比,培养基筛选等方面对石栗组织培养进行了探索。结果表明诱导愈伤组织的主要因素是细胞分裂素和生长素的比值。TDZ对愈伤组织诱导丛生芽有抑制作用。诱导愈伤组织的最适培养基为:1/2MS+ZT 2 mg/L+NAA 0.5 mg/L+氯化胆碱2 mg/L,诱导愈伤组织丛生芽的最适培养基为1/2MS+6-BA 1 mg/L +IBA 0.1 mg/L。影响诱导愈伤组织的主要因素是细胞分裂素和生长素的比值。

Abstract:

The aims were to establish the technique system which was about the tissue culture of Aleurites moluccana, and then choose the media which was the most appropriate for calli induction and differentiation. The results showed that, the ratio of the cytokinin and the archusia was the primary factor that acts on the calli induction. TDZ inhibited the induction that from the calli to the axillary buds. The best media for calli induction was: 1/2MS+ZT 2 mg/L+NAA 0.5 mg/L+choline chloride 2 mg/L. The most suitable media for inducing axillary buds was 1/2MS+6-BA 1 mg/L +IBA 0.1 mg/L. The ratio of the cytokinin and the archusia was the important factor for calli induction.
