伊维菌素人工抗原的制备及鉴定

Preparation and Identification of the Artificial Antigen of Ivermectin

摘要: 为制备伊维菌素人工完全抗原,通过对伊维菌素进行化学修饰,引入活性基团羧基,合成具有半抗原结构特征的伊维菌素半琥珀酸酯。然后采用NHS法将半抗原与牛血清白蛋白(BSA)和鸡卵清蛋白(OVA)偶联,制备人工免疫原IVM-BSA和检测原IVM-OVA,经SDS-PAGE和紫外光谱扫描鉴定。用人工免疫原IVM-BSA免疫Balb/c小鼠,间接竞争ELISA法测定抗血清效价。结果发现:偶联物IVM-BSA和IVM-OVA的偶联比分别为14.1:1和14.7:1,免疫小鼠抗血清效价达到1:12800,结果说明成功制备出人工完全抗原并且具有良好的免疫原性。

Abstract: The purpose of this experiment was to prepare complete artificial antigens of ivermectin. The synthesis of ivermectin half succinate which had hapten structure was performed through introducing active carboxyl group. Artificial antigens, named IVM- BSA and IVM- OVA, were obtained by means of coupling ivermectin with macromolecular carriers, such as bovine serum albumin (BSA) and ovalbumin (OVA), using NHS method. The identification of artificial antigen was conducted by SDS-PAGE and UV scanning methods subsequently. Balb/c mice were immunized with complete antigen IVM-BSA and antiserum titer was tested by indirect competitive ELISA method. The results showed that the coupling ratio of IVM-BSA and IVM-OVA were 14.1:1 and 14.7:1 respectively. The antiserum titer was 1:12800. It came to the conclusion that IVM-BSA and IVM-OVA were synthesized successfully and had satisfied immunogenicity.
