甜叶菊同源四倍体与二倍体基因组差异分析

Analysis of Genetic Variation in Autotetraploid and Diploid Stevia Rebaudiana Bertoni

摘要: 为探究甜叶菊二倍体经染色体加倍后得到四倍体的过程中,基因组序列的变化情况,采用ISSR分子标记技术对1 个甜叶菊二倍体及其5 个同源四倍体株系的遗传差异进行分析。ISSR 结果显示:从60 个ISSR 引物中筛选出多态性强、重复性好的5 个引物进行扩增,共扩增出34 条谱带,其中有19 条呈多态性,多态性占55.88%;6 个甜叶菊株系间Nei's 基因多样性指数范围在0.0059~0.2778 之间;利用聚类分析,发现CK与5 个四倍体最先分开,这说明聚类分析可以很好的将甜叶菊二倍体和同源四倍体区分开,1 和2、4 和5 两两株系最先聚在一起,这表明2 组株系各自间遗传相似度高。甜叶菊不同倍性间遗传差异较大,同一倍性不同株系间也存在一定的遗传差异。

Abstract: In order to research the changes of genomic sequence after Stevia rebaudiana Bertoni diploid chromosome doubling, ISSR molecular marker technique was used to analyze the genetic variation between 1 diploids and 5 autotetraploids of Stevia rebaudiana Bertoni. 5 primers with good polymorphic and steady bands were selected from 60 ISSR primers, and 34 bands were amplified totally. 19 of 34 bands were polymorphic, the polymorphic rate was 55.88%. The genetic diversity index were 0.0059-0.2778 in 6 Stevia rebaudiana Bertoni lines. The UPGMA dendrogram showed that CK and 5 tetraploids separated firstly, which explaining that Stevia rebaudiana Bertoni diploids and autotetraploids were separated well by cluster analysis. 1 and 2 lines, 4 and 5 lines clustered together firstly, which indicating that the genetic similarity coefficient of the two groups were high. There was significant genetic variation between autotetraploid and diploid of Stevia rebaudiana Bertoni, and different lines with the same ploidy also had genetic variation.
