猴腿蹄盖蕨孢子萌发和成苗影响因素初探

The Effects of Spores Germination and Planting Rate of Athyrium multidentatum

摘要: 为了提高猴腿蹄盖蕨孢子人工培养成苗效率,以猴腿蹄盖蕨孢子为外植体,通过在组织培养中加入不同种类激素和NaH2PO4及活性炭,探讨了猴腿蹄盖蕨孢子萌发和成苗影响因素。结果表明:20mg/LGA3处理成熟孢子5min,孢子萌发速度加快;NAA、2,4-D、BA、KT四种激素中KT促进孢子萌发效果最好,BA和NAA次之。1/2MS,MS两种培养基中,1/2MS培养基孢子萌发速度快于MS培养基;NaH2PO4浓度以200~300mg/L可促进幼苗迅速生长,形成大量丛生苗;培养基中添加2%活性炭有利于小苗的生长。

Abstract: In order to raise efficiency of planting seedling by artificial spore culture of Athyrium multidentatum, in this article, spore of Athyrium multidentatum was taken as the explants, and used different categories of hormone, NaH2PO4 and AC during the period of tissue culture. The result showed that the methods of plant tissue culture were used for effects 5 levels of time of treatment of GA3,4 hormone(NAA, 2,4-D, BA, KT) of 5 levels concentration, 2 cultural ground substances,5 levels of GA3 concentration applying on spores germination; the effects 6 levels NaH2PO4 and 5 levels AC on planting rate were studied. The results showed that the 20mg/LGA3 most suitable treatment time was 5 min, the most suitable hormone was KT, BA and NAA take second place, the most suitable medium was 1/2MS on spores germination; The most suitable concentration of NaH2PO4 was 200~300mg/L, the most suitable AC was 2% in cultural ground substances on planting rate.
