改良CTAB-LiCl法提取枣总RNA体系的建立

摘要: 本试验以枣树田间枝条的枝皮及组培苗为材料,应用改进的CTAB法进行了总RNA的提取。针对枣组织中富含多糖多酚的特点,改进了CTAB法,采用β-巯基乙醇和PVP去除酚类,KAc及无水乙醇去除多糖,LiCl沉淀过夜的方法提取得到的RNA,条带整齐、清晰,A260/A280介于1.8~2.0之间,A260/A230均大于2.0,证明得到的RNA纯度、完整性和产率均较高,蛋白、多糖及酚类等去除较彻底。结果表明,本试验建立的改良CTAB-LiCl法适于进行枣总RNA的提取

Abstract: In this paper, the branches of Chinese jujube and its plantlets were used as test materials to establish the optimum system of isolating total RNA. Because Chinese jujube are rich in polysaccharides and polyphenol compounds, the CTAB method were improved by added polyvinylpyrrolidone(PVP) and β-ME to remove polyphenol compounds, added KAc and ethanol to remove polysaccharides and added LiCl to deposit RNA. The bands of the isolated RNA were regular and clear. The A260/A280 of total RNA were between 1.8~2.0 and A260/A230 were more than 2.0. That proved the method could extract high-purity and highly-complete RNA at a high productivity and could remove protein, polysaccharides and polyphenol compounds completely. The result showed that the improved CTAB-LiCl method is suitable to isolate total RNA of Chinese jujube.
