叶籽银杏trnS-trnG序列测定与分析

Sequencing and analysis of trnS-trnG on Ginkgo biloba var. epiphylla Mak

摘要: 利用叶绿体基因trnS-trnG基因间隔区的通用引物,扩增并分析了不同地区的14株叶籽银杏和2株银杏的trnS-trnG基因间隔区序列,结果表明,14株叶籽银杏和2株银杏的trnS-trnG序列长度均为1005bp,A+T的含量为62.19%—62.39%。各样品之间变异小,14株叶籽银杏和2株银杏共有23个可变位点,1个信息位点,这些变异都以碱基的置换发生。除这些变异位点外的核苷酸序列完全相同,说明各样品序列之间具有高度同源性。YZ1变异位点较多,与其它样品的同源性较低。所测序列Blast检验后发现叶籽银杏trnS-trnG序列与银杏的相似度最高,苏铁类次之。用DNASTAR软件对14株叶籽银杏和2株银杏的trnS-trnG基因间隔区序列进行同源性和差异性分析,建立了系统进化树。

Abstract: The cpDNA trnS-trnG intergenic spacer sequence of 14Ginkgo biloba var. epiphylla trees and 2 Ginkgo biloba trees were amplified and analyzed successfully.The results indicated that the size of trnS-trnG sequences was 1005bp and the content of A+T was 62.19%—62.39%.There were 23 variable sizes and one information size in this 14 Ginkgo biloba var. epiphylla trees and 2 Ginkgo biloba trees.There were little difference among the trnS-trnG sequences.YZ1 had more variable sizes and lower different areas with other samples.All the sequences through Blast found that it had a highly similar with Ginkgo biloba and cycas another.DNASTAR software was used to analysis indentity and diver gence and construct phylogenetic tree based on all the trnS-trnG sequences of 14Ginkgo biloba var. epiphylla trees and 2 Ginkgo biloba trees.
