Research of the T-DNA Introduced to Watermelon and Molecular Marker Via the Stigma Smeared Method

摘要:借鉴拟南芥蘸花进行外源DNA导入的方法,通过柱头涂抹进行西瓜T-DNA导入和分子标记研究。结果表明:品种ZXG01078坐果率最高,且有变异苗产生,是较为理想的介导品种;卡那霉素筛选处理西瓜种子的最佳浓度500~700mg/L,不同品种间有差异;卡那霉素筛选处理西瓜叶片的最佳部位为嫩叶或幼叶,最佳浓度4000~8000mg/L,不同品种间差异不明显。处理后代中无心苗和连体苗的稳定产生,说明外源基因已进入并干扰了其正常的生长发育过程。

Abstract:

Abstract:From the method of the foreign DNA introduced to Arabidopsis thaliana via flowers dip, we used stigma smear method to transfer T-DNA into watermelon and research T-DNA molecular marker. The results showed the ideal tranform specie is ZXG01078 with the highest fruit setting rate and the most deviant seedlings. The best concentration of the watermelon seeds treated by kanamycin is 500~700mg/L with differences among the species; the best concentration of the watermelon leaf treated by kanamycin is 4000~8000mg/L, and the best position is spire leaf or youngleaf with no difference among the species. The steadily variation appearing of no pointing and twinborn seedling indicated the normal growth interfered by foreign DNA in the progeny.
