川麦冬组织培养技术研究

摘要:

摘 要: 以川麦冬根的嫩基部为外植体,对其无菌苗的诱导、增殖及不定根的诱导进行了初步的研究。结果表明,用75%酒精20s+0.1%HgCl210min消毒效果最好。在以MS+6-BA2.0mg/L+NAA0.2mg/L的培养基中进行的不定芽诱导生长和继代培养,诱导和增殖效果较好,丛生芽的一代诱导率为80%,二代以后丛生芽诱导率为100%。在继代至第四代时丛生芽的平均数目达到6.24个,完全可以满足快速繁殖的需要。麦冬组培苗不定根的最适培养基为1/2MS+NAA0.5mg/L,生根率达100%,田间移栽2周后成活率可达到100%,且幼苗生长良好。

Abstract:

Abstract: With the tender stem segments with partial root of Ophiopogon Japonicus (L.f.)Ker-Gawl as the explants, the induction of sterile plantlet, formation of re-generated plantlet, differentitation and growth of adventitious root were studied. The results showed that disinfecting with 75% alcohol for 20s and 0.1% HgCl2 for 10 min had the best disinfection effect. The effect of bud-inducing and multiplication was better on the medium MS+6-BA2.0mg/L+NAA0.2mg/L,and the inducing rate of asepsis plantlet was 80%, the inducing rate of clumpy buds was 100%.The average of clumpy buds was 6.24 on the propagation of fourth and could perfectly meet the need for rapid multiplication. The inducing rate of adventitious root with the best medium 1/2MS+NAA0.5mg/L was 100%. When transplanted in the farm after two weeks, the survival rate of plantlets was 100%, and the plantlets were vigorous.
