麻栎的组织培养与快速繁殖

摘要:

以麻栎幼苗茎段为外植体,以MS为基本培养基,研究了不同种类和不同浓度生长调节物质对麻栎组织培养的影响。结果表明,芽诱导最佳培养基为MS+6-BA1.0 mg?L-1,培育21天左右,腋芽可长至1~2 cm,展叶3~4片,发芽率达83.3%;芽增殖最佳培养基为MS+6-BA1.0 mg?L-1+NAA0.02 mg?L-1,增殖系数为4.6;最佳生根培养基为1/2MS+NAA0.5 mg?L-1,培养4周后生根率可达87%。

Abstract:

Effects of plant hormones to the tissue culture of Quercus acutissima were studied by means of seedling stems as explants and MS as basic medium. The results showed that the best medium for the bud induction was MS +6-BA1.0 mg?L-1. The bud was as long as 1 to 2 cm with 3 to 4 leaves after three weeks, and the germination rate was 83.3%. The best medium for the bud proliferation was MS +6-BA1.0 mg?L-1 + NAA0.02 mg?L-1, and the proliferation rate was 4.6. The best medium for rooting was 1/2MS + NAA0.5 mg?L-1, and the rooting rate was about 87% after four weeks.
