Analysis of Genetic Diversity of SPF Leghone Using Microsatellite Markers

Abstract: 【OBJECTIVE】Five microsatellite markers were used to analysis two closed colony of SPF Leghone by calculation of the mean genetic heterozygosity, polymorphism information content in order to provide information for STR database of SPF Leghorn chickens.【METHOD】PCR products of Five microsatellite markers detected with ABI3100-Avant genetic analyzer and analyzed with software DataCollection and GeneMapper 3.1.【RESULTS】The results showed that 20 alleles were detected of 5 microsatellite loci,the average alleles of each loci was 4.The average heterozygosity of the two chicken colony was 0.6211 and the average of the polymorphism information content was 0.6663,which indicated high polymorphic of the 5 microsatellite loci in SPF Leghone.The allele 180bp of adl176 loci was specific to population A and conspicuous difference of the two alleles of 188bp、191bp were detected between two closed colony,which implied ideal primer for line identification.【CONCLUSION】The mojority of microsatellite markers are high polymorphism and we can find specific ones for genetic monitor
