Identification of the Pathogen of Radish Wilt in Beijing Suburb and Factors Affecting its Growth

Abstract:

A new radish (Raphanus sativus L.) disease happened in Beijing suburb in 2009. The main symptom of the disease is that the infected radish grows stunt and it’s leaves become yellow. We collected the typical diseased radish plants to isolate the pathogen, and identified the pathogen according to the morphology and the sequence of the internal transcribed spacer region of the ribosomal DNA (ITS), and also studied the major affecting factors of the pathogen in order to provide the basis to study controlling measures and to screen the resistant germplasm resources and select resistant cultivars to the disease. The results were showed as follow: the pathogen of the disease was identified as Verticillium dahliae; The optimal temperature for the pathogen growth was 20-25℃; The optimal pH was 6-8; The pathogen can utilize many kinds of carbon sources and nitrogen nutrient very well, ammonium sulfate was the best nitrogen nutrient and soluble starch was the best of carbon sources for its growth. This was the first report about Verticillium wilt of radish in China.
