AnalEST-SSR Analysis and Primer Screening of Turnip Mosaic Virus (TuMV) Resistance in Chinese Cabbage

Abstract:

In order to study turnip mosaic virus (TuMV) in Chinese cabbage, ESTs in Chinese cabbage and resistant ESTs in Brassica were searched and SSRs were screened. Primers were designed according to the sides sequence and were screened using the genome DNA of one pair of TuMV resistant and susceptible materials of Chinese cabbage as templates. The results showed that a total of 166 (16.2%) SSRs were screened from 132 Chinese cabbage ESTs and 895 resistant ESTs of Brassica, among which there were six kinds of nucleotide repeat motifs, the dinucleotide (33.7%) and trinucleotide (50.0%) repeats were the main types, proportions for other repeat types were much small. 196 primers were designed according to the sides sequence of the above SSRs, among which 156 (79.6%) were amplified successfully, and 35 primers were amplified polymorphic in the two materials. Therefore, SSR primers designed from Brassica resistant EST sequences could be used in marking of TuMV resistant genes of Chinese cabbage.
