Different Plant Growth Regulator Treatment on the Regeneration of Upland Cotton Cotyledon Sections

Abstract:

To establish a high frequency regeneration system for the gene transformation of cotton, cotyledon nodes of three cotton varieties were acted as the explants, regeneration of cotton cotyledon section was modified and optimized to, especially the cotyledon node rooting was discussed. The results showed that: For bud induction, the best inductive medium suitable for ‘Baimian 1’ was MSB + 1.0 mg/L 6-BA and 0.5 mg/L IBA; the MSB + 2.5 mg/L 6-BA + 2.5 mg/L KT was suitable for ‘Baimian 2’. The MSB medium supplemented with 2.0 mg/L 6-BA was appropriate for ‘Coker 312’. Each explant could obtain 3 to 4 buds, and most of the buds could be elongate. For rooting, MSB+0.5 mg/L NAA medium was utilized.
