Effect of Different Explants and Phytohormone on Callus Induction of Camellia oleifera

Abstract:

The study aimed to provide the technical reference to rapid propagation and biotechnology breeding of Camellia oleifera. Effect of different explants and phytohormone on callus induction of Camellia oleifera was studied by using the asepsis seeding of leaf、stem segments and shoot tip as the explants. Combination of 2,4-D and KT were the best phytohormone for the inducting of callus. In the condition of MS basic medium added with 0.5 mg/L 2,4-D and 1.0 mg/L KT, the callus induction rate were higher when used leaf and stem segments as the explants, while In the condition of MS basic medium added with 0.5 mg/L 2,4-D and 2.0 mg/L KT, the callus induction rate were higher when used leaf and stem segments as the explants. In the three kinds of explants which were selected, leaf was the easiest for inducting, and induction rate could reach 100%. From the induction rate and the amount of plant hormones, leaf is the best explants of the three kinds of explants while inducting callus.
