Establishment of in vitro Regeneration System for Processing Tomato

Abstract:

Cotyledons of processing tomato cultivar ‘Yaxin 98-1’ and ‘Liger 87-5’ were used as explants and cultured on the medium with different mitogen (ZT or 6-BA) and auxin (IAA or NAA) to study their effect on callus induction and bud differentiation. The results showed that callus could be induced in all kinds medium, but there were obvious differences on callus configuration and bud differentiation. The effect of ZT was better than that of 6-BA, and IAA was better than NAA on bud induction. The optimum medium for bud differentiation was MS+ZT 0.5 mg/L+IAA 0.2 mg/L. The bud differentiation of cultivar ‘Yaxin 98-1’ was better than that of cultivar ‘Liger 87-5’. The regenerated buds could develop normally after roots were induced on 1/2MS+IAA 0.1 mg/L.
