Genetic Transformation of Chitinase and β-1, 3-Glucase Gene to Rubber Tree

Abstract:

The aim was to enhance the resistance of powdery mildew of rubber tree with genetic engineering. 15090 anther callus of high yield, large-scale spread, elite variety ‘CATAS 7-33-97’ was used as receptor in this study, bean chitinase and tobacco β-1,3-glucanase gene were transformed into Hevea mediated by Agrobacterium, 1386 resistant somatic embryos were differentiated, then proliferated by secondary embryogenesis, 277 were successful. PCR results indicated that 49 embryos were positive, with 0.33% transformation efficiency. Bean chitinase and tobacco β-1, 3-glucanase gene were transformed into Hevea mediated by Agrobacterium, with 0.33% PCR positive efficiency. It laid a good foundation for improving the powdery mildew resistance by genetic engineering in Hevea.
