Isolation and Identification and Phylogenetic Analysis of Yeast Strains from the Intestinal Tract of Mink

Abstract: In order to get the mink dedicated beneficial bacteria and provide a theoretical basis for the study fur animal probiotic preparations. Two strains of yeasts were isolated and purified from the intestines of healthy mink named JM1 and JM2. Morphological observation, physiological and biochemical tests and 26S rDNA gene sequence analysis, and a phylogenetic tree were constructed. Meanwhile, these two bacteria’s growth and security were comprehensive identification. The traditional identification methods and 26SrDNA D1/D2 sequence analysis results were showed that: JM1 was identified as Saccharomyces cerevisiae strain and JM2 was Pichia fabianii strain; The growth result was that: the growth speed of two strains were faster in 14-30 h and the growth speed were stabilized in 30-40 h; sugar content in the culture medium of the two strains Baume degrees began to rapidly decline at 12 h, and showed an increasing trend after 26 h. Safety test results were showed that: 2 strain had no toxicity on mice, and the mice grew at the fastest speed at the 2.73×107cfu/mL dose.
