Effect of Temperature and pH on Digestive Enzyme Activities of Misgurnus anguillicaudatus

Abstract: The effects of temperature and pH value on digestive enzyme activities of adult Misgurnus anguillicaudatus were investigated. Folin-phenol, 3,5-dinitrosalicylic acid and p-nitrophenol were prepared to determine the activities of protease, amylase and lipase in M. anguillicaudatus intestine and hepatopancrea. The optimum temperatures of protease, amylase and lipase in intestine were 40℃, 45℃ and 35℃, respectively. The optimum temperatures of protease, amylase and lipase in hepatopancreas were 30℃, 40℃ and 35℃, respectively. All the optimum pH values of three digestive enzymes in intestine or hepatopancreas were 7.2. The A/P values both in intestine and hepatopancreas were greater than 1. The results suggested that the dietary of M. anguillicaudatus could be increased with the rise of water temperature at the appropriate range for M. anguillicaudatus cultivation. M. anguillicaudatus was more like herbivorous, and more capable digestibility for carbohydrate. The formulation of M. anguillicaudatus diet might be more proportion of carbohydrate, it could fulfill the requirement of nutrition and be more commercial.
