Effect of Cryoprotectant and Freezing Rate on Semen Cryopreservation of Breeder Cock

Abstract: In this study, it was aimed to improve the freezing effect of the frozen semen from Hy-Line Brown breeder cock. Therefore, the effects of cryoprotectant and freezing rate on their semen cryopreservation were also investigated respectively. In the first experiment, both DMA and DMSO were applied as cryoprotectants, and their concentration were 6%, 10%, 15% respectively. The results showed as following: in the group of DMSO (10%), the percentage of total motility of sperm was 37.4%,the percentage of total recovery rate of sperm was 50% and the viability rate of sperm was 58.3%. In the second experiment, three cooling rates were designed totally and compared with one another. The results showed as following: in the best cooling rate group, total motility rate of sperm reached to 44.3%, recovery rate reached to 53%, and morphologically abnormal sperm rate and acrosome integrity rate were 41.9% and 42.9% respectively. Finally, the effect of frozen Breeder cock semen was also evaluated by their artificial insemination (24.9%). Collectively, it can be concluded that combination of 10% DMSO with the cooling rate being treated by the temperature from 5℃ to -35℃at 7℃/min, and then from -35℃ to -120℃ at 9℃/min,initiate the best inseminated effect of their freezing semen.
