Development of Swida wilsoniana Microspore and Its Effect on Anther Callus Induction

Abstract: To establish an in vitro regeneration protocol for Swida wilsoniana ‘Xianglin G1’, an elite clone, calluses were induced from its anthers with different development stages of microspores. The flower morphology at different development stages of microspores and the endogenous hormones in leaf such as GA, IAA, ZR, and ABA were also conducted to comprehensively understand the influence of microspores at different stages on the callus induction of anthers. The results showed that callus induction of anthers was dependent on the development stage of microspores. The anthers with single-nucleus microspores had higher induction rates than those with bi-nucleus microspores, tetrad microspores, and pollen mother cells. The callus induction of anthers correlated with the concentration of GA, IAA, ZR, and ABA in leaf, and higher ABA content in leaf significantly inhibited the callus induction of anthers.
