Cloning and Sequence Analysis of ShSAP1 promoter from sugarcane

Abstract: Promoter play an important role in transgenic breeding, discovering of active promoters, especially stress-specific and organ/tissue-specific promoter is of great interest to manage the concerns associated with gene expression efficiency. According to the genome sequence of ShSAP1, a member of stress associated proteins (SAPs), a 1243 bp promoter region of ShSAP1 was isolated from sugarcane genome by genome walking. Sequence analysis found the promoter region contained core promoter elements and other ciselements like drought and other stress response elements, MYB/MYC recognition and binding site, light responsive elements and tissue specific expression elements, which indicated ShSAP1 promoter might be a stress-specific and tissue-specific promoter. The function of ShSAP1 promoter still needs further study.
