Abstract: To improve the selection accuracy for exogenous insect resistant gene in cotton breeding, nontransgenic insect-resistant cotton varieties and the transgenic insect-resistant cotton varieties with single & two exogenous insect resistant genes were used to establish and optimize the PCR method for detection of exogenous insect resistant gene. Non-transgenic cotton lines and transgenic cotton lines were detected by Btspecific primers and SAD1 primers (an endogenous gene in cotton), the results showed that the individual in 40% of the transgenic plant lines has no expected amplicon, 8.0% individual in the non- transgenic lines GANG 197 has expected amplicon. This method can be applied to cotton breeding for exogenous gene detection and improves selection accuracy.
