Study on Priming Treatments of Seed Dormancy Breaking in Phellodendron amurense Rupr.

Abstract: Seed of Phellodendron amurense Rupr presented different degrees of dormancy which varied with provenance and year. Conventional cold stratification was only partially effective in breaking dormancy of treated seeds. In order to solve the problem of low rate of seed germination of Phellodendron amurense Rupr, explore the most effective way to break seed dormancy, different chemical- initiators were used to treat the seeds, and then combined with 4℃or 8℃cold stratification. In this paper, the results showed that soaked with 6-BA, KNO3, PEG 6000 could promote seed germination. In addition, pretreated with 1000 mg/L GA3, 80 mg/L NAA, 400 mg/L 6-BA, 1.0% KNO3, 20 g/L PEG6000 that combined with 4℃cold stratification for 30 d could break seed dormancy more effectively and make the germination rate increased from 13.50% to 91.00% , 94.00%, 97.00%, 94.50%, and 83.00% respectively. The priming could effectively break the seed dormancy and promote seed germination of Phellodendron amurense Rupr and shorten the time of cold stratification.
